Study On Chemical Constituents Of Aphanamixis Polystachya And Design,Synthetic And Anti-PEDV Studies Of Phenanthridium Derivatives

First part of this thesis studied on the chemical constituents of Aphanamixis polystachya(Wall).Almost thirty compounds were isolated,identificated eighteen new compounds inclouding a nortriterpenoid,two triterpenoid and five steroid compounds.In the second part,12 derivatives were designed and synthesized from lycorine,then studied on whether and how the derivatives keep PEDV from multiplying.This study suggest that most derivatives had anti-PEDV activity,and some derivatives anti-PEDV by down-regulating the host Hsc70 protein expression.Chapter 1:Introduced the significance of study on the chemical constituents of Aphanamixis polystachya.Isolated thirty compounds from the twigs and leaves of Aphanamixis polystachya,eighteen compounds were identified,including eight new compounds(including a nortriterpenoid,two triterpenoid,five steroid compounds),ten known identified compounds inclouding one triterpenoid,four steroids and five sesquiterpenoids.Other compounds is about triterpenoids,norterpenoids and sterides.Chapter 2:Listed the reasons for selecting phenanthridium derivatives for anti-PEDV activity research.Designed and synthesized 12 compounds(1-12)from lycorine according to previous studies on the structure-activity relationship of phenidine compounds,then studied the anti-PEDV activity about the 12 compounds.Results suggested that some derivatives had anti-PEDV activity,among which compounds 1,2 and 12 inhabit the PEDV replice efficiently,and their inhibitory activities are 2.9,2.3 and 5.2 times of the control group respectively.Further studies on the mechanism of anti-PEDV suggest that the activity of some compounds is related to the heat shock homologous protein(Hsc70)in host cell,results show that by incubation host cells with compounds 2 or 12,PEDV replication were inhabited,and Hsc70 expression in host cells were reduced,it has been proved a positive correlation.
