ObjectiveTo investigate the effect of compound Ganoderma Lucidum Rhodiola preparation on the immune function of immunosuppressed mice induced by cyclophosphamide,and its effect of anti-fatigue ability in mice.Methods1.Immunosuppressive mouse model was prepared by cyclophosphamide.Two doses of Ganoderma lucidum Rhodiola compound preparation were used to intervene immunosuppressed mice.The Ganoderma lucidum Rhodiola compound preparation extracted by different extraction methods was used to intervene immunosuppressed mice.2.Anatomize the thymus and spleen of normal mice,immunosuppressed mice and drug-treated mice.Calculate the thymus index and spleen index of the mice,collect the peripheral blood of each group of mice,and count the leukocyte in peripheral blood.3.Prepare spleen cell suspension of each group of mice,the proliferation ability of T cells and B cells were detect by the method of MTS/PMS,and the IL-6 level,IL-10 level,TNF-? level and IFN-? level in spleen cell culture medium were detect by ELISA.the expression of CDa+T cells and CD8+T cells in spleen cell culture medium was detected by flow cytometry,and the ratio of CD4/CD8 was calculated.4.the mice treated with the enzyme-treated compound Ganoderma lucidum Rhodiola preparations for 28 days,the exhaustive swimming time of drug-treated mice and normal mice were compared.5.eyeballs were taken to collect blood,urea nitrogen was detected by urease method,lactate dehydrogenase content and lactate content were detected by micro-enzyme method,and MDA was detected by thiobarbital method.6.The liver and gastrocnemius muscles of mice were dissected,and the liver glycogen and muscle glycogen storage of the mice were detected by colorimetry.Results1.Compared with cyclophosphamide group(CTX),the spleen index,peripheral blood leukocyte count,T lymphocyte proliferation rate,B lymphocyte proliferation rate and TNF-? expression level significantly increased in Ganoderma lucidum compound preparation(CWO)low dose group,Ganoderma lucidum rhodiola enzyme extraction compound preparation(CEO)low dose group,CWO high dose group and CEO high dose group(P<0.05)2.Compared with cyclophosphamide group(CTX),the expression levels of IL-6 and IL-10 in CEO high-dose group and CWO high-dose group were significantly increased(P<0.05);the expression levels of IFN-? in CEO low-dose and CEO high-dose group were significantly increased(P<0.05);the CD4/CD8 significantly increased in CWO high-dose group,CEO low-dose and CEO high-dose group(P<0.05).3.Compared with the CWO high-dose group,the IL-10 and TNF-a levels in the high-dose CEO group significantly increased(P<0.05).Compared with the CWO low-dose group,the IFN-y levels in the low-dose CEO group significantly increased(P<0.05).Compared with the Control group,the CWO low-dose group,the CWO high-dose group,the CEO low-dose group and the CEO high-dose group were not statistically significant on organ index,leukocyte count,T lymphocyte proliferation ability,cytokine level and CD4/CD8 level.4.Compared with the control group,the swimming time,blood lactate dehydrogenase,hepatic glycogen and muscle glycogen of the Ganoderma lucidum Rhodiola compound preparation group significantly increased(P<0.05).5.Compared with the control group,the contents of blood lactate,urea nitrogen and malondialdehyde in the mice of Ganoderma lucidum Rhodiola compound preparation group significantly decreased(P<0.05).Conclusion1.Compound Ganoderma lucidum Rhodiola preparation can enhance the immune function of cyclophosphamide-induced immunocompromised mice.2.The compound Ganoderma lucidum Rhodiola preparation after enzyme treatment is better than the direct water extraction compound Ganoderma lucidum Rhodiola preparation3.The compound Ganoderma lucidum Rhodiola preparation can enhance the anti-fatigue ability of mice...
