In recent years,with the improvement of human living standards,has led to many obesity problems.Among the many factors leading to obesity,in addition to excessive energy intake and a small amount energy consumption,human body Abnormal expression of genes involved in production and degradation of adipocyte is also an important factor leading to obesity.Many scientists have attempted to provide a better theoretical basis for the treatment of clinical obesity-related diseases by studying the regulatory mechanisms of genes involved in adipocyte differentiation.The DHHC family of proteins is an extremely conservative protein that was first discovered in yeast and has 24 DHHCs in mammals.Due to the specific cysteine residue structure of these proteins,the current study of DHHC is mainly used as a membrane protein to transfer proteins between plasma membranes by palmitoylation modification.Since DHHC interacts with fatty acid-palmitic acid,we speculate that DHHC may be related to adipocyte metabolism.To test this hypothesis,we first detected the expression of the DHHC gene in adipose tissue and then screened for DHHC4 and DHHC5 by differential differentiation of adipocytes,which have an effect on the differentiation of adipocytes.The DHHC5 study found that:it is involved in the early transcriptional regulation of adipocyte differentiation,and further studies have shown that it is involved in the phosphorylation of Akt and S6K in the insulin signaling pathway.Finally,we constructed Rosa-Ert2-cre Dhhc5f/fmice,and the same results as in the 3T3-L1 cell experiment were obtained by separating the SVF experiments.The conclusion is that DHHC5 affects the early differentiation of adipocytes through the insulin signaling pathway.These provide us with a new perspective for the study of DHHC function,and provide a feasible experimental method for the study of DHHC protein in fat differentiation,and also provide a theoretical basis for the treatment of obesity.
