ObjectiveTo investigate the expression of Tim-3 in blood Treg-enrich population of patients with AR,by stimulating and blocking the signal pathway of Ti-m-3/Tim-3L on CD4+CD25~+FoxP3~+Treg of peripheral blood of patients with allergic rhinitis(AR),and to observe the changes of Tim-3/Tim-3L on the proportion of CD4~+CD25~+FoxP3~+Treg of CD4~+T cells in AR patients,as we l-l as the changes of cytokines related to the secretion of Treg,to study o-n the value of regulating the Tim-3/Tim-3L in immunological mechanisms of AR.MethodsThis experiment is divided into three parts:part one:The peripheral b-lood of 3ml from 30 patients with allergic rhinitis(AR)and 25 healthy co-ntrols(HCs)were collected.The proportion of CD4~+CD25~+FoxP3~+Treg and Tim-3~+Treg in AR group and HCs group was detected by flow cytometry,and the relationship between Tim-3 expression and Treg was analyzed;p-art two:The peripheral venous blood of 3ml was collected from 30 patien-ts with AR.After Tim-3 blocker was given,to detect the percentage of Tr-eg and IL-10~+Treg before and after adding reagent by flow cytometry;par-t three:The peripheral venous blood of 3ml was collected from 25 patien-ts with AR.After Galectin-9 was given,to detect the percentage of Treg a-nd IL-10~+Treg before and after adding reagent by flow cytometry.Results1.The percentage of Treg in peripheral blood of patients with AR[1.20%(0.51%~1.79%)]was lower than that of healthy controls of HCs[5.02%(4.70%~5.67%)].(Z=-6.339,P<0.01)2.The expression of Tim-3 on the surface of Treg in AR patients[11.10%(6.96%~16.21%)]was higher than that in healthy controls of HCs[3.34%(2.71%~3.82%)].(Z=-5.570,P<0.01)3.The expression of CD4~+CD25~+FoxP3~+Treg cells in AR patients was negatively correlated with the expression of Tim-3 on the surface.(r=-0.763,P<0.05)4.The percentage of Treg in peripheral blood of AR patients after blo-ck[1.72%(0.98%~2.26%)]was not significantly changed compared with that before blocking[1.33%(0.68%~2.05%)].(Z=-1.617,P=0.106)The pe-rcentage of IL-10 secreted by Treg cells in peripheral blood of AR patient-s after block[1.93%(1.41%~3.14%]was higher than that before block[1.16%(0.75%~1.72%)].(Z=-4.784,P=0.000)5.The percentage of Treg in peripheral blood of AR patients after act-ivation[2.61%(2.38%~2.87%)]was significantly higher than that before activation[2.29%(2.12%~2.67%)].(Z=-2.960,P=0.03)The percentage of IL-10 secreted by Treg cells in peripheral blood of AR patients after activa-tion[1.75%(1.52%~2.81%)]was higher than that before activation[1.14%(0.97%~1.78%)].(Z=-4.372,P<0.05)Conclusions1.The expression of Treg with allergic rhinitis was down-regulated,w-hile that of Tim-3~+Treg was up-regulated.2.After administration of Tim-3 blocker,the percentage of IL-10~+CD4~+CD25~+FoxP3~+Treg cells in peripheral blood of patients with allergic rhinitis(AR)was increased.3.After the administration of activator Gal-9,the percentages of CD4~+CD25~+FoxP3~+Treg cells and IL-10~+CD4~+CD25~+FoxP3~+Treg cells in patients with allergic rhinitis(AR)were increased.
