Objective: To observe the changes of CD8+ T cell subsets and their relationship with EBV-DNA load before and after treatment in patients with nasopharyngeal carcinoma.Methods: Flow cytometry was used to detect the percentage of CD8+ T lymphocyte subsets in peripheral blood of 10 healthy people and 11 patients with nasopharyngeal carcinoma before and after treatment.The initial T cells(CD45RA+CCR7+)and terminal differentiation effector T cells were used.(CD45RA+CCR7-),central memory T cells(CD45RA-CCR7+),and effector memory T cells(CD45RA-CCR7-)accounted for the percentage of total lymphocyte counts,respectively,and analyzed the changes of cell subsets.The EBV-DNA concentration in peripheral blood of patients with nasopharyngeal carcinoma was determined by EBV-DNA test kit,and the CD8+ T cell subset was analyzed for its association with viral load.Results: 1.In the comparison between the healthy group and the pre-treatment lymphoid subgroup of nasopharyngeal carcinoma patients,the proportion of primary cells(CD45RA+CCR7+)in the nasopharyngeal carcinoma patients was lower than that in the healthy group,the difference was statistically significant(P<0.05).The proportion of central memory cells and effector memory cells(CD45RA-CCR7+,CD45RA-CCR7-)subgroup was higher than that of healthy group,the difference was statistically significant(P<0.05);effector cells(CD45RA+CCR7-)subgroup There was no significant change in the proportion of the healthy group.Before and after treatment of nasopharyngeal carcinoma,the proportion of primary cells(CD45RA+CCR7+)subgroup and central memory cells(CD45RA-CCR7+)subgroup was lower than that before treatment,the difference was statistically significant(P<0.05);The CD45RA+CCR7-)subpopulation was significantly higher than the previous one,and the difference was statistically significant(P<0.05).3.In the EBV-DNA high load and low load group comparison,the EBV-DNA high load group The proportion of effector memory cells in the lower load group was low,and the difference was statistically significant(P<0.05).There were no statistical differences in the comparison of the other three cell subpopulations.Conclusion: The proportion of lymphocyte subsets is significantly different between NPC patients and healthy people,the initial cell ratio is decreased,and the proportion of central memory cells and effector memory cells is increased.The proportion of lymphocyte subsets changed significantly before and after treatment,the ratio of initial cells to central memory cells decreased,and the proportion of effector cells increased.By detecting changes in the proportion of cell subpopulations and providing more information for assessing immune function in patients,lymphocyte subsets are expected to be an indicator for assessing the immune status of patients.
