Objective:Investigating growing environment and resources distribution of Clerodendranthus spicatus(Thunb.)C.Y.Wu at Xishuangbanna,and general confirming its growing environment and resources distribution;collecting samples,at the same time some samples are bought from different herbal medicine dealers and used to research quality standard.Rosmarinic acid,Caffeic acid,Orthosiphol F,Ursolic acid,Kiganic acid are regarded as quantity indicators,testing analysis methods on standard products,and testing Principal components in samples by HPLC to establish the components'testing method,and provide an evidence on quality standard of Clerodendranthus spicatus(Thunb.)C.Y.Wu.Orthogonal testing optimizes extraction technology of Clerodendranthus spicatus(Thunb.)C.Y.Wu,HPLC tests qualitative and quantity indicators,and confirm principal components on quality standard;calculating content and extraction rate in principal components to build the best method of extraction technology on principal components.Methods:Generally fix growing environment and resources distribution of Clerodendranthus spicatus(Thunb.)C.Y.Wu by studying references,inspecting samples collected and actual visit,and collect and conduct samples from different growing areas,different reaping time and different medicinal parts.According to People's Republic of China Pharmacopoeia(Four,2015),HPLC method tests and confirms key contents in Clerodendranthus spicatus(Thunb.)C.Y.Wu.Orthogonal testing optimizes extraction technology:the samples from the same year reflow extraction in different conditions,firstly,in different temperatures;secondly,in different hours;thirdly,in different liquid ratio;and the last is different ethanol concentration,after the solution chilling,filtering and spindling it into dry extract at 80?,and exactly weight dry extracts,and using pure water to dissolve the dry extracts into 250ml's capacity bottles,filter the solution in 0.45?m micro-porous and tests the principal contents in samples.Count extraction rate of Rosmarinic acid in standard curve equation;Count Extraction rate of Caffeic acid in standard curve equation;Count Extraction rate of Orthosiphol F in standard curve equation.According to People's Republic of China Pharmacopoeia(Four,2015),to test TLC.According to People's Republic of China Pharmacopoeia(Four,2015),to test water content,leachate,total ash and acid insoluble ash in samples.Results:Generally confirm growing environment and resources distribution of Clerodendranthus spicatus(Thunb.)C.Y.Wu at Xishuangbanna;its medicinal parts are divided into stem and leaf,purple flower and white flower are mainly species;plant origin and growing environment are the same with China Flora.HPLC testing shows that peaking reserving time of Rosmarinic acid and Caffeic acid are the same with the standard products.Content in Rosmarinic acid is 0.096~1.824(?g/ml),R~2=0.9997;content in Caffeic acid is0.145~5.8(?g/ml),R~2=0.9999;content in Orthosiphol F is 0.4~4(?g/ml),R~2=0.9998;Content in Ursolic acid is 0.32~6.08(?g/ml),R~2=0.9983;content in Kiganic acid is 0.32~6.08(?g/ml),R~2=0.9989.Optimized chromatographic condition:Chromatographic column:ZORBAX SB-C18(4.6×250mm 5?m);mobile phase:A Phosphoric acid water(1%phosphoric acid)-B Acetonitrile(gradient elution);colunm temperature:30?;flow rate:1ml/min;wavelength:203nm,time:82min?Optimized extraction condition of Rosmarinic acid:temperature:83?;time:1.8h;liquid to material ration:50ml/g;ethanol concentration:17%?Optimized extraction condition of Caffeic acid:temperature:83?;time:2h;liquid to material ration:50ml/g;ethanol concentration:67%?Content standard:Rosmarinic acid is 7.49%,Caffeic acid is 2.76%.Diterpene F is not in examination area,it is not regarded as a quantity indicator,andUrsolic acid and Kiganic acid are not tested.Rosmarinic acid mean content in different conditions in purple stem and white stem:4.95mg,0.84mg.Caffeic acid mean content in different conditions in purple stem and white stem:1.34mg,1.05mg.Terpenes and Organic acid can be shown by TLC.Water content is 10%,leachate is 12%,total ash is 5%,and acid insoluble ash is 2%.Conclusion:Mean temperature of Clerodendranthus spicatus(Thunb.)C.Y.Wu at Jinghong is 20~22?,and the rainfall is 1200~1900mm.Resources distribution is in Institute of Medicinal Botany,Gasa town,Menghai and Mengla County.Content tested in Rosmarinic acid and Caffeic acid may be regarded as qualitative and quantifiable products.Rate extracted in different conditions is a kind of reliable and the best extraction method.Content of Rosmarinic acid and Caffeic acid in purple stem is much higher than white stem.Samples tested by TLC presents good results,TLC examined can be considered as a standard.Content tested in Water,leachate,total ash and acid insoluble ash can be regarded as a standard.
