In this dissertation, Dandelion as a traditional Chinese herbal medicine was studied and the quality control of Dandelion was further investigaged by modern analytical technology. In addiontion to the determinations of the content of Dandelion as stated in Chinese Pharmacopoeia published in 2005, in this work, the content of caffeic acid, chlorogenic acid, ferulic acid was simutanously determined, and the fingerprint of Dandelion was obtained, furthermore, the content of water, total ash, acid insoluble ash, water extracts and heavy metals in Dandelion was also determined. Based on the results obtained in this work, a large number of valuable information and data can be provided for the revision of the new quality standards in Chinese Pharmacopoeiam. The main research in this work was as follows:Objective: To establish a satble, feasible and comprehensive quality standard of Dandelion.Method: (1) The identification and qualitative analysis of Dandelion were carried out with character identification, microscopic identification and TLC.(2)The method for the measument of the content of caffeic acid, chlorogenic acid, ferulic acid was established used high capillary electrophoresis (HPCE) as the analytical technology.(3) UV-VIS was used to establish an analytical method for the determinations of the content of total flavonoids in Dandelion(4) High performance capillary electrophoresis (HPCE) was used to study the fingerprint of Dandelion obtained from different areas and different varieties in Henan province.Then the results was evaluated by the Similarity Evaluation System for Chromatographic Fingerprint of TCM (2004A) developed by the Pharmacopoeia Committee of China, and the standard fingerprints of Dandelion in Henan were established.(5)Based on the regulations in Chinese Pharmacopoeia published in 2005 Appendix, the content of water, total ash, water extract and heavy metals in Dandelion was determined, and then the detection items can be added and improved accordingly. Results: (1) Refering to "the Chinese Pharmacopoeia" (in 2005 version), the upper formation solution of the ethyl acetate - formic acid - water (7:2.5:2.5) was the developing solvent. The sample has the same color of fluorescent spots under the uviol lamp (254 nm) to inspect compare to the position of standards of caffeic acid, chlorogenic acid.(2) Extraction method and content determination condition of coffeic acid, chlorogenic acid , ferulic acid and fingerprint in dandelion were established according to single factor experiment and orthogonal test and by the selections of the testing conditions such as buffer system, buffer solution PH value, buffer solution density, voltage, examination wave length and so on.(3) In this work, the water, total ash, acid insoluble ash, water extract of hot dip method, the water extracts of infusion and heavy metals in dedanlion were tested in order to prefect its Inspection project.Conclusion:(1) A capillary electrophoresis method for simultaneous determination three organic acids of caffeic acid, chlorogenic acid and ferulic acid in dandelion was accurate, reliable, stable and feasible.(2) The method of UV-VIS for total flavonoids in dandelion was accurate, reliable, simple and easy.(3) The method of combinating HPCE Fingerprint with determination of a variety of active ingredients in dedanlion in Henan can be used to evaluate Henan dedalion. The method was scientific, accurate, fast and energy saving, environmental protection and a green chemical analysis method. Based on the results obtained in this research, the quality of Henan Dedalion can be better controlled, and this work can provide a certain conbutions to greatly develop the industry of dedalion products and to develop the economy of Henan province. |
