Colon cancer is one of the most prevalent malignancies in the world,obesity will increase the risk of colon cancer and the number of obese people is increasing by an unhealthy diet in many countries and regions.As a result,the risk of colon cancer increases,which seriously affects people's physical and mental health.Studies have shown that genistein as an estrogen can stimulate the apoptosis of tumor cells.However,genistein has rarely been studied for the prevention of colon cancer.In this experiment,female SPF grade Kunming mice were selected as the experiment.Firstly,high-fat diets containing different doses of genistein and normal feeds were separately fed to establish a high-fat model(the weight of mice in the high-fat group is more than 20%in the normal group).Then,based on the established high-fat model,AOM/DSS method was used to induce the colon cancer model in mice.Finally,the prevention and molecular mechanism of genistein in mouse colon tumor was explored.Through the evaluation of mouse colonic cancer disease activity index(DAI),it was found that the DAI value of mice in high-fat groups was greater than that of normal mice.After feeding genistein,the DAI value of mice became smaller,and DAI(900 mg/kg)in high dose group<DAI(450 mg/kg)in medium dose group<DAI in low dose(225 mg/kg)<DAI in induction group,which indicated that genistein can prevent colon tumor.The mouse serum testing results showed that the TG value significantly increased,but the TC and LPS content did not change significantly in AOM/DSS induction group.Feeding in the genistein group,the TG,TC,and LDL-C levels were reduced,and the HDL-C levels were increased,indicating that genistein has the effect of lowering blood lipids in mice.Histopatho logical section results showed that the structure of the glands in the colon of the high-fat mice induced group was completely disordered,while the structure of the colon tissues of mice fed the genistein group was substantially intact.The RT-PCR results of colon cancer related genes showed that the intake of genistein can reduce the expression of COX-2,TNF-? and FRAT1 mRNA and increase the expression of PTEN mRNA.Western blotting indicated that genistein could inhibit the expression of PI3K,AKT and Bcl-2 protein in the colon of mice,promote the expression of FOX03,Bax and Caspase8 proteins,and the inhibit the growth of colon tumor in mice.In this experiment,the preventive effect of genistein on colon cancer in mouse was analyzed at the molecular level,and the preventive effect was positively correlated with the intake of genistein.The preventive mechanissms may be related to genistein's regulation of mouse colon cancer gene and protein expression.
