Dendrobium officinale Kimura et Migo,is an entire grass with perennial epiphytic herb and belongs to Dendrobium Sw.,orchidaceae,with a focus on many provinces in the southwestern region.It belongs to the more important Chinese medicinal materials,and has the effects of various aspects such as stomach,heat and lungs.In the actual analysis process,an in-depth analysis was carried out on the preparation procedure and specific structure of the stem polysaccharide,and the key analysis contents are as follows:1.Separation and purification of polysaccharides from Dendrobium candidumIn the actual analysis,the water extraction method is selected to prepare the desired polysaccharide material,and the enzyme and the Savage method are used again to eliminate the protein present therein.Finally,Sephadex G-75 chromatography can be used in the analysis.Two types of substances required.In the actual analysis,electrophoresis and chromatography are used to identify the purity parameters,and the DOP1 component is determined to belong to a homogeneous substance,while DOP2 is attributed to a special mixture.2.Structural analysis of polysaccharides from Dendrobium candidumIn the analysis process,the viscosity method was used to determine the mean molecular weight parameter of DOP1 was 3.78×104.After the corresponding acid hydrolysis procedure,DOP1 was determined by the matching chromatograph to obtain the corresponding monosaccharide structure,namely mannose and galactose.The specific molar ratio parameter is:2.221:1.375:2.010:2.322:1.After the corresponding procedures such as sodium periodate oxidation and 13CNMR,spectrum analysis,it is presumed that there is a corresponding 1→6 glycosidic bond.3.Antioxidant activity of DOP1 and DOP2For the corresponding removal of ·OH,O2-· experimental analysis,the final analysis of the polysaccharide DOP1And DOP2 has a corresponding activity,and will increase with the increase in concentration.
