ObjectiveBy establishing PCOS rat model,we analyse the relationship between nesfatin-1 and serum reproductive endocrinology hormones.And we also investigate the expression of nesfatin-1 mRNA and protein in PCOS and control rats,in order to explore the scientific basis of nesfatin-1 in the pathogenesis of PCOS.MethodsThirty 6-week-old adolescent female SD rats were randomized into two groups,the PCOS group and the control group.PCOS was successfully induced with oral dosing of letrozole at 1.0 mg/kg daily for 21 days.The 15 rats in the control group were given placebo.Body weight measurements were taken at a frequency of every day.Estrous cycles were monitored by vaginal smears that were performed throughout the entire duration of the experiment.The ovarian morphology was observed by HE staining;the levels of serum hormone were measured by electrochemiluminescence assay;the serum nesfatin-1 was measured by ELISA;fasting blood glucose was measured by hexokinase kinase assay;the expression of nesfatin-1 were analyzed by Immunohistochemistry?Real-time PCR and western blot in the hypothalamus,stomach and ovary of rats.Results(1)The body weight,ovarian weight,and ovaries quotiety in the PCOS group was significantly higher than in the control group(P<0.05).(2)The levels of LH,T,FBG,FSI and HOMA-IR in the serum of PCOS group were significantly higher than those of the control group,while the levels of Nesfatin-1,FSH,E2 and P were lower than those in the control group.Serum Nesfatin-1 was negatively correlated with body weight,LH,T,FSI and HOMA-IR,and positively correlated with FSH and P.(3)Compared with the control group,the ovarian tissue of PCOS group showed more early follicular follicles and cystic dilatation follicles,and the number of granulosa cells decreased and the number of corpus luteum decreased significantly.(4)Compared with the control group,the positive staining of hypothalamus,stomach,and ovary cells in PCOS group was significantly decreased by immunohistochemical staining.(5)The expression of NUCB2/nesfatin-1 in hypothalamus,stomach,and ovary of each group was detected by qRT-PCR and W-B method,suggesting that the expression of NUCB2/nesfatin-1 in the PCOS group was lower than that in the control group(P<0.05).Conclusion(1)The rat model of PCOS was successfully induced with oral dosing of letrozole.(2)The expression of NUCB2/nesfatin-1 was significantly decreased in the PCOS group,and nesfatin-1 has a negative correlation with body weight,LH,T,FSI,and HOMA-IR,whereas it has a positive correlation with FSH and P.This study provides a new insight in the pathogenesis of PCOS,and nesfatin-1 maybe a useful marker for the diagnosis of PCOS.
