Objective:In this study,we hope to express both tumor-associated macrophages(TAMs)labeled antigen CD68 and alpha/beta hydrolase 5(ABHD5)in gastric cancer and adjacent normal tissues.The situation was tested,and the relationship between the expression of CD68 and ABHD5 and clinicopathological parameters of gastric cancer was further analyzed in combination with clinical analysis.The roles of the two in the occurrence,development,and prognosis of gastric cancer and the correlation between the two were preliminarily discussed.Method:60 medical records of patients with gastric cancer undergoing surgical resection in the First Affiliated Hospital of Bengbu Medical College and archived paraffin blocks were selected as study subjects.The time range was from January 01,2014 to December 31,2014,and 30 cases of cancer were selected.Using normal tissues as controls,the expression of TAMs labeled antigens CD68 and ABHD5 in gastric cancer and adjacent normal tissues was detected by immunohistochemical method.Whether the difference between the two expressions was statistically significant was statistically significant.The relationship between clinical pathological parameters such as the age of onset,gender,pathological grade,clinical stage,lymph node metastasis,depth of invasion,etc.,was expressed in the tissue,and the correlation between CD68and ABHD5 expression in gastric cancer tissues was analyzed by correlation.During the whole experiment,the prognosis of gastric cancer was analyzed by telephone or outpatient follow-up,and independent prognostic factors were analyzed by COX risk factors.Results:1.The positive expression rate of TAMs labeled antigen CD68 in gastric cancer tissues was 65.00%(39/60),which was significantly higher than that of normal tissues adjacent to cancer 26.67%(8/30).The difference was statistically significant(?2=11.779,p=0.001).CD68 expression correlated with clinical TNM staging(?2=7.959,p=0.005),lymph node metastasis(?2=6.901,P=0.009),invasiveness(?2=5.833,p=0.016),and pathological grading(?2=0.028,p=0.866),gender(?2=0.533,p=0.465),and age(?2=0.060,p=0.807).2.The positive rate of ABHD5 expression in gastric cancer tissues was 60.00%(36/60),which was significantly higher than that in paratumor normal tissues(30.00%,9/30).The difference was statistically significant(?~2=7.200,p=0.007).The expression of ABHD5 was associated with pathological grade(?2=4.318,p=0.038),TNM stage(?2=4.051,p=0.044),degree of infiltration(?2=5.275,p=0.022),and lymph node metastasis(?2=0.545,p=0.460),gender(?2=0.191,p=0.662),age(?2=1.358,p=0.244).3.In gastric cancer tissues,correlation analysis found that there was a positive correlation between the expression of CD68 and ABHD5 in gastric cancer tissues(r=0.328,p=0.010).4.After three years of follow-up,the overall survival rate(OS)of cases with strongly positive CD68 expression was significantly longer than that of the negative cases(p<0.05);Similarly,the OS with strongly positive ABHD5 expression was lower than that of negative patients(p<0.05),and the difference was statistically significant.Univariate analysis showed that pathological grading,clinical stage,lymph node metastasis,depth of invasion,and expression of CD68 and ABHD5 all had an impact on prognosis.Multivariate results showed that ABHD5 and clinical stage were independent prognostic factors.Conclusions:1.TAMs was highly expressed in gastric cancer tissues,and the expression level was down-regulated in normal gastric cancer,and its expression level decreased with the increase of clinical stage of gastric cancer.ABHD5 lower the expression in normal tissue adjacent to carcinoma,the expression in gastric cancer tissue,and its expression level with the increasing of tumor pathological grading and clinical staging and heighten,prompt TAMs and expression of ABHD5 is closely related to the proliferation,invasion and metastasis of gastric cancer.2.In gastric cancer tissues,there was a positive correlation between the expression of TAMs labeled antigen CD68 and ABHD5,and the specific mechanism needs further elucidation.3.The expression of ABHD5 is an independent prognostic factor of gastric cancer,which contributes to the evaluation of metastasis,invasion and prognosis of gastric cancer.
