ObjectiveResearch the Effects of Nesfatin-1 on Cholesterol side chain cleavage enzyme and P450 Aromatase by Human Luteinized Granulosa Cells.MethodsThe screening of 20 cases of in vitro fertilization and embryo transfer(IVF-ET)of reproductive medicine,Affiliated Hospital of Binzhou Medical University.All patients received long-term treatment of ovulatory follicle stimulating and NG were classified into two groups:PCOS group and normal control group(n=10).The cumulus cells of human luteinized granulosa cells in mature oocytes oocyte retrieval.Add serum containing medium(containing 10%fetal bovine serum)after purification in vitro,quantity statistics and adjustment of granulosa cells before the SS in 24 pore plate 2X105/ml inoculation density,and then add the medium with cincentration nesfat 1X10-9 M without serum for 24 h.media were collected to measure P450scc concentration and P450arom using chemiluminescence detection.Results1.The expression of Cholesterol side chain cleavage enzyme in PCOS group and normal control group increased with the same concentration nesfatin-1(1X10-9M)in ovarian luteinized granulosa cells cultured in vitro.At the same concentration,the expression level of Cholesterol side chain cleavage enzyme in ovarian luteinized granulosa cells in PCOS patients was significantly lower than that in normal controls(p<0.05).2.The expression of P450 Aromatase in PCOS group increased with the same concentration nesfatin-1(1X10-9M)in ovarian luteinized granulosa cells cultured in vitro(p<0.05).There was no significant difference in the level of nesfatin-1 between the normal control group in vitro(p<0.05).ConclusionNesfatin-1 can promote the secretion of follicular lutein particles,increase the concentration of P450scc and Nesfatin-1 it can promote the secretion of Nesfatin-1 in PCOS,aromatase secretion,also has no significant effect in the normal control group.
