Objective:To investigate the effects of different concentrations of Nesfatin-1 on estradiol and progesterone by human luteinized granulosa cells in vitor.Methods:Screening of 20 cases received in-vitro fertilization-embryo transfer(IVF-ET) from department of reproductive medicine of Binzhou Medical University Hospital.All patients given long follicle-stimulating therapy for ovulation induction were divided into two groups:PCOS group (n=20) and normal control group(n=15). Human luteinized granulosa cells obtained from the cumulus granulosa cells around the mature oocyte in oocyte's retrival day, joined containing serum culture medium(DMEM+10%fetal bovine serum) in vitor 48 hours after purification.The numbers of granulosa cells were counted and adjusted,press the density of 2X105/ml vaccination in 24 hole plate,and then cultured with medium plus various cincentration of nesfatin-1(1X10-7M?1X10-9M?1X10-11 M?0) without serum for 24 hours. The media was collected to measure the concentration of estradiol and progesterone by luminescence detection.Results:1. Luteinized granulosa cells were cultured in vitro with different concentration of nesfatin-1 (1X10-7M?1X10-9M?1X10-11 M?0) for 24 hours.we found that:PCOS group and normal control group luteinized granulosa cells generate E2 level increased with the nesfatin-1 concentration increasing. And in different concentration of nesfatin-1, PCOS group of luteinized granulosa cells E2 production levels were significantly lower than the normal control group (P<0.05).2. Luteinized granulosa cells were cultured in vitro with different concentration of nesfatin-1(1X10-7M?1X10-9M?1X10-11 M?0) for 24 hours.we found that:PCOS group luteinized granulosa cells cultured in vitro with the nesfatin-1 generate P level was significantly increased than basic condition (P<0.05). Normal control group luteinized granulosa cells cultured in vitro with the nesfatin-1 generate P level compared with basic condition was no significant difference (P> 0.05).And in different concentration of nesfatin-1, PCOS group of luteinized granulosa cells P production levels were significantly lower than the normal control group (P<0.05).Conclusion:Nesfatin-1 can promote the secretion of estradiol in human follicular luteinized granulosa cells, and the effect with nesfatin-1 concentration increasing; Nesfatin-1 can promote the secretion of progesterone secretion in PCOS group, no obvious effect on progesterone secretion in normal control group, which may be through with the period in which the granulosa cells and whether there are FSH.
