| Objective: to compare the difference of GLP-1R expression on human islet cells between three glycemic states of normal glucose,impaired glucose tolerance and diabetes,and analyze the effect of common clinical factors on GLP-1R expression,in order to better guide the clinical application of GLP-1 analogues in the treatment of type 2 diabetes.Method: the Fujian provincial hospital provided samples of human pancreatic tissue that had been surgically removed from January 2008 to December 2013.A total of 74 cases of complete clinical data were obtained by retrospective analysis.According to the different glycemic definition process,these human pancreatic tissue samples were divided into three groups: diabetes group(16 cases),glucose tolerance group(29 cases),and non-diabetic group(29 cases).The positive expression was measured by immunohistochemical method,and the positive expression was evaluated by full quantitative and semi quantitative count respectively.Result: 1、 Baseline Information: the statistical results showed that there was no significant difference in sex,age and average BMI between the three groups(P > 0.05).The duration of the three groups and the history of treatment were statistically different.Further stratified analysis showed that there was a significant difference in the duration of the three groups(P < 0),so the duration of the disease was the interference factor of GLP-1R expression difference in different blood glucose levels.2、 Immunohistochemical Results: there were positive expressions in the three groups of pancreatic tissues,of which GLP-1R was stained with cytoplasm and cell membrane,and the positive expression was brown.The statistical results showed that there was no significant difference in the total and semi quantitative expression of the positive expression of the three groups of GLP-1R and islet cells(P > 0.05).The Pearson correlation test coefficient of GLP-1R and islet cell volume in the group was 5.41,and the results were statistically significant(P < 0.01).There was no statistical difference between the expression of GLP-1R and the duration of diabetes(P > 0.05).Conclusions: there was no significant difference in the expression of GLP-1R on pancreatic islet beta cells in human pancreatic tissue under different blood glucose conditions,but GLP-1R was positively correlated with islet cells.There is no significant difference in the expression of GLP-1R in the duration of diabetes,suggesting that the length of diabetes may not affect the expression of GLP-1R. |
