Objective: Cholangiocarcinoma is the second most common type of primary liver cancer worldwide which originates from the epithelium of biliary.The high mortality rate and poor prognosis of CCA are associated with early invasion,widespread metastasis,and the lack of an effective therapy.EIF3 d is a gene that plays an important role in the translation initiation.This important function leads eIF3 d have currently considered to be one of the reasons causes numerous tumors disease.However,the functional role of eIF3 d in cholangiocarcinoma cancer remains unclear.Our study mainly focused on the effects of eIF3 d gene expression in cholangiocarcinoma cells and to investigate the effects of eIF3 d on proliferation,migration,and cell cycle,in vivo.To investigate whether the eIF3 d gene can affect the growth of cholangiocarcinoma cells and the role in cholangiocarcinoma,presumably its role in the development of cholangiocarcinoma.Methods: 1.By constructing lentiviruses and transfecting,we obtained stable cholangiocarcinoma HCC-9810 and RBE cell lines with low expression of e IF3 d gene.2.In vitro cell experiments such as colony formation assays,MTT assays,and transwell assays validated the effects of eIF3 d gene on proliferation and migration of cholangiocarcinoma cells in vitro.Flow cytometry was used to examine the effect of eIF3 d on the cell cycle of cholangiocarcinoma cells.Western-blot(over-imprinting assay)was used to detect changes in the expression of related proteins.3.A nude mouse model was constructed to investigate the effects of the eIF3 d gene on tumor growth in animals.Results: By transfection with lentivirus,we obtained stable cholangiocarcinoma HCCC-9810 and RBE cell lines with low expression of eIF3 d gene.In subsequent cell experiments,Knockdown of e IF3 d in HCC-9810 and RBE cells significantly inhibited cell proliferation and colony formation in vitro.Flow cytometry analysis indicated that depletion of eIF3 d led to cell-cycle arrest in the G2/M phase.In vivo experiments suggest that the size of the tumor in the eIF3 d knockdown group is much smaller than that of the control group.Conclusion: This study explored the role of eIF3 d gene in the growth and metastasis of cholangiocarcinoma cells and found that the eIF3 d gene plays an important role in the proliferation and migration of cholangiocarcinoma.eIF3 d can promote the proliferation and migration affect the cell cycle progression of cholangiocarcinoma.Our study may provide a potential site and new method for the diagnosis and treatment of cholangiocarcinoma.
