ObjectiveStudy the expression of WIF-1 protein in the extrahepatic bile duct carcinoma and analyze the relationship of WIF-1 protein expression and clinical pathological characteristics.To study the mechanism of 5-aza-2-deoxycytidine(DAC),a methylation inhibitor,effect on the invasion of cholangiocarcinoma cells(TFK-1).MethodsDetect the expression of WIF-1 protein in 38 cases of extrahepatic bile duct carcinoma tissue and 15 cases of chronic cholangitis tissue by immune histochemical method(SP),and analyze the relationship of WIF-1 protein expression and clinical pathological characteristics.Transwell method was used to detect the invasion of cholangiocarcinoma cells under different concentrations(0?10?20?40?80?160 ?mol/L)of DAC.Western-blot method was used to detect the expression of WIF-1 protein.Results1.Positive expression rate of WIF-1 protein was 21.0% in the 38 cases of extrahepatic bile duct carcinoma and positive expression rate of WIF-1 was 100% in the 15 cases with chronic cholangitis tissues,there were significant difference(P <0.05).Expression of WIF-1 protein was related to the level of differentiation of extrahepatic bile duct carcinoma,WIF-1 protein expressed in the well-differentiation of extrahepatic bile duct carcinoma tissue,poorly differentiated rare expression in extrahepatic bile duct carcinoma tissue.The expression of WIF-1 was not related to sex,age,tumor size and lymph node metastasis(P>0.05).2.DAC inhibited cholangiocarcinoma cells in concentration-dependent manners.Protein of WIF-1 didn't express on the cholangiocarcinoma cells,but Protein of WIF-1 reexpressed on the cholangiocarcinoma cells after dealing with DAC.Conclusions1.Expression of WIF-1 protein in extrahepatic bile duct carcinoma were higher than that in chronic cholangitis tissues.Expression of WIF-1 protein in the extrahepatic bile duct carcinoma correlated with the level of differentiation,there were no obvious relationship with the sex,age,tumor size,lymph node metastasis.Which explained WIF-1 protein lower expression may associated with the development of extrahepatic bile duct carcinoma.2.DAC inhibited invasion of cholangiocarcinoma cells.Maybe this dued to the methylation of DAC.
