Objective :To study the hypoglycemic effect of three different dosage forms(decoction,dipping agent and powder)of Rhizoma Coptidis on the fasting blood glucose and regulation of gastrointestinal motility and gastrointestinal hormones of diabetic rats.Exploring the relationship between gastrointestinal motility,gastrointestinal hormones and blood glucose and the possible mechanism of different formulations of Rhizoma Coptidis.And then initially clear the best use of Rhizoma Coptidis(dosage form)and reasonable and effective dose,to guide the clinical rational use of berberine to prevent diabetes and its complications to provide new ideas.Method:120 male SD rats were randomly divided into normal control group(n = 10)and diabetic model group(n=110).The rats were fed with normal diet and high fat and high glucose diet for 15 days.After fasting can not help water for 12 hours,,according to 40 mg / kg dose intraperitoneal injection STZ modeling.Detecting fasting blood glucose after 72 hours,FGB?16.7mmol / L were diabetic rats.The rats were randomly divided into 8 groups and10 groups in each group.They are respectively dipping agent size dose group,decoction size group and powder size dose group.The next day of grouping,All the rats were weighed and given a corresponding concentration of 10ml·kg-1 rats.Continuously given a gavage for 2 weeks.Observing he general state of experimental animals daily.Every 3 days to detect a weight,,weekly detection of fasting blood glucose.After 2 weeks of intragastric administration,fasting can not help water for 12 hours,giving a gavagae of charcoal paste,D-xylose for an hour one after another.Then collecting blood from the femoral artery and executing the experimental animals.Lastly,detecting the gastrointestinal motility and preparing specimen.According to the kit method to detect the indicators.Results:1.Three different formulations of Rhizoma Coptidis can control the weight loss of diabetic rats and reduce the fasting blood glucose level.Hypoglycemic effect in the high dose of decoction group,high dose of immersion agent group,the powder group is better.And there were no significant difference to compare with Metformin.The hypoglycemic effect was no significant difference between the high dose group.The low dose group had the best effect of lowering podwer.2.There was no significant effect of three different formulations of Rhizoma Coptidis on serum insulin level,but three forms can significantly perfect insulin resistance.Between the groups,the most effective groups are three high-dose group and small dose of powder group.3.There was a significant gastrointestinal motility disorder in diabetic rats with STZ,which showed an increase in gastric residual rate and small intestinal propulsion rate.Three different formulations of Rhizoma Coptidis can improve STZ-established diabetic rats gastrointestinal motility disorders,reduce the residual rate of the stomach,increase the rate of small intestine.The curative effect was best in the high dose group of impregnant and powder,it is superior to Metformin in improving gastric emptying,and has no difference in improving intestinal propulsive rate.The curative effect of decoction group was poor,and the improvement rate of intestinal propulsion was not as good as Metformin.4.Diabetic model rats had gastrointestinal hormone disorder,which showed that the levels of GAS and MOT were significantly increased and the level of SS was significantly decreased.The dosage of different dosage forms of Rhizoma Coptidis could promote the secretion of GAS,MOT,SS and GLP-1,and increase the level of hormone in tissue homogenate.Among them,the high dose group of powder had the best effect on improving the levels of GAS,MOT and SS.The high dose group of decoction had no obvious effect on GAS and MOT,The dipping agent had no obvious effect on GAS.The combined effect of powder with the best group,followed by dipping agent,decoction effect is poor.5.The D-xylose in the serum of diabetic rats was significantly higher than that in the normal group.Three different formulations and different dosage forms of Rhizoma Coptidis can inhibit the absorption of D-xylose in the small intestine.Among the three formulations,the best effect was in the high dose group of decoction.It was no evident difference with Metformin.6.The levels of GLP-1 were significantly decreased in the tissue bomogenate of diabetic rats.The different forms of Rhizoma Coptidis can improve the expression of GLP-1 in small intestine tissue,the effect is equivalent to Metformin.The best effect was the high dose group of dipping agent.Conclusions:1.Different dosage forms of Rhizoma Coptidis can control the weight loss of diabetic rats,improve insulin resistance,reduce fasting blood glucose.,the powder is the best.2.Different dosage forms of Rhizoma Coptidis can regulate the secretion of gastrointestinal hormones GAS,MOT and SS,improve gastrointestinal motility disorder and inhibit the absorption of D-xylose in small intestinal.Among the three formulations,the powder had the best effect on Improvement of gastrointestinal motility,the decoction had the best inhibitory effect on intestinal absorption.3.Different dosage forms of Rhizoma Coptidis could promote the secretion of GLP-1 and increase the level of GLP-1 in the small intestine,and the best effect was obtained by three kinds of dosage forms?The best was the dipping agent.4.On the one hand,the hypoglycemic mechanism of Rhizoma Coptidis is related to the promotion of GLP-1 secretion and insulin sensitivity.On the other hand,it may be related to the improvement of gastrointestinal motility and related hormones,inhibition of small intestinal glucose absorption,and increase the utilization rate of glucose.5.The mechanism of improving gastrointestinal motility may be related to lowering the level of blood glucose,improving the secretion of gastrointestinal hormones GAS,MOT and SS related to gastrointestinal motility,regulating hormone imbalance and enhancing the sensitivity of hormones.In conclusion,different formulations of Coptis could reduce the fasting blood glucose level,to achieve the same powder of low dose and high dose Decoction and impregnating effect.The powder can reach the same effect with the large dose of the decoction and the dipping agent at a small dosage.The utility of the powder can be used to save medicinal materials,and is convenient to carry.It is is superior to the decoction and impregnant.For patients with diabetes mellitus with obvious gastrointestinal motility disorder,the impregnation agent and powder are more ideal than the decoction.
