There are two parts in the thesis.The first part is theorizing Rhizoma Coptidis and its different formulation in the treatment of diabetes.The second part is the scientific research on the effects of different formulation of Rhizoma Coptidis on the blood glucose and the activities of intestinal disacchridases in type 2 diabetic rats.Theoritic explorationIntroducing the understanding of diabetes mellitus,the theory of treating diabetes mellitus with Rhizoma Coptidis,and the records and research of different formulation of Rhizoma Coptidis in treating diabetes mellitus,from the perspective of traditional Chinese medicine,tutor's views and research.Scientific researchObjective: By using the diabetic rats as animal model,observing the effects of different formulation of Rhizoma Coptidis on the blood glucose and the activities of intestinal disacchridases in type 2 diabetic rats,exploring the best formulation of Rhizoma Coptidis and the most effective dose,and to explore its possible mechanisms and targets,to guide the clinical rational use of prevention and treatment of diabetes and provide references.Methods:(180 20)g male SD rats were randomly divided into normal control group(n = 10)and diabetic model group(n = 110).The normal control group were fed with normal diet;the diabetic group were fed with high fat and high sugar diet for half a month.Fasting for 12 hours,rats were intraperitoneally injected STZ(40mg/kg)to build the model of type 2 diabetes and then fed with normal diet.72 h later,select the rats FGB more than 16.7mmol/L as diabetic rats,then divided into 8 groups randomly:low-dose group and high-dose group of decoction,low-dose group and high-dose group of dipping,low-dose group and high-dose group of powder,normal model group and positive control group.The feeding time was two weeks.At the end of the experiment to detect the blood glucose of rats and the related indexes.Results:(1)Each group of Rhizoma Coptidis could relieve the symptoms of polydipsia,polyuria,polyuria and emaciation in diabetic rats.Compared with the model group,the high-dose of decoction?The high-dose and low-dose group of dipping and the high-dose and low-dose group of powder can significantly control the weight loss in diabetic rats(P<0.05);(2)The high-dose group of decoction?dipping?powder and the low-dose group of powder can significantly lower fasting blood glucose in diabetic rats(P<0.05),and there was no significant difference between groups(P>0.05).From the trend of the effect,the high-dose group of powder was better;(3)Compared with the model group,the serum insulin content of each group of Rhizoma Coptidis have no statistically significance(P>0.05).The high-dose group of decoction?dipping?powder the low-dose group of powder can significantly improve insulin resistance in diabetic rats(P<0.05),and there was no significant difference between groups(P>0.05).From the trend of the effect,the high-dose group of powder was better;(4)The high-dose and low-dose group of decoction?The high-dose and low-dose group of dipping can inhibit the activities of intestinal lactasein in diabetic rat(P<0.05);and there was no significant difference between groups(P>0.05);The high-dose group of decoction?dipping?powder can significantly inhibit the activities of intestinal maltase(P<0.05);and there was no significant difference between groups(P>0.05);The high-dose group of dipping can significantly inhibit the activities of intestinal sucrase in diabetic rat(P<0.05);Conclusion:(1)the high-dose group of decoction?dipping?powder and the low-dose group of powder can significantly lower fasting blood glucose and improve insulin resistance in diabetic rats.From the trend of the effect,the high-dose group of powder was better;(2)The high-dose groups were obviously better than the low-dose group in inhibiting the activities of intestinal disaccharidases in diabetic rats.From the trend of the effect, the high-dose group of dipping was better;(3)The mechanism of lowering fasting blood glucose may have something to do with improving insulin resistance and inhibiting the activities of intestinal disaccharidases.
