Exosomes are nanograde membrane-bound vesicles secreted from most cell types through the fusion of multivesicular bodies with plasma membranes.Some of these exosomes are well defined,and are known to have immunomodulatory properties as well as play critical roles in intercellular communications.In this study,we characterized the role of exosomes secreted by Trichomonas vaginalis in immune responses and analyzed its effect on tumor cell proliferation and metastasis.Objective:To explore the role of T.vaginalis exosomal mediated cervical epithelial cells,prostate epithelial cells immune response and the proliferation and metastasis of cervical and prostate cancer cells.Methods: T.vaginalis positive samples collected from the department of obstetrics and gynecology were cultured in vitro and the exosomes identified by TEM and nanoparticle tracking analysis were extracted.The uptake of T.vaginalis exosomes by Ect1?RWPE1?Siha and PC3 cells was detected through labeling with fluorescent dye PKH67.After exosome stimulation,the production of interleukin IL6 and IL8 in Ect1?RWPE1?Siha and PC3 were investigated in vitro using enzyme-linked immunosorbent assay(ELISA),q PCR was used to detect m RNA levels of several interleukins in cells.The proliferation of Siha and PC3 cells affected by exosomes was evaluated with MTT.Finally,the changes in the invasion of Siha and PC3 cells were detected through wound healing assay and transwell assay,and the changes in the related proteins about epithelial mesenchymal transformation(EMT)were observed.Results: The exosomes derived from T.vaginalis were successfully isolated and characterized,and T.vaginalis exosomes labeled with PKH67 dye could be taken up by host cells Ect1?RWPE1?Siha?PC3.The production of both IL6 and IL8 were increased in Ect1 and RWPE1 through incubation with T.vaginalis exosomes,but not in Siha and PC3.Furthermore,exosomes stimulation of tumor cells had no clear effect on the proliferation of Siha and PC3 cells.However,it could induce the enhancement of migration and invasion mediated by EMT.Conclusion: T.vaginalis exosomes act on cervical epithelial cells and prostate epithelial cells,causing inflammatory response.It can also lead to increased cervical cancer cells and prostate cancer cells metastasis,and EMT is one of the mechanisms causing this change.
