ObjectiveGastric cancer remains to be one of leading causes of cancer related death all of the world.Mitochondria is the energy source of eukaryocyte cells.UCP2(uncoupling protein-2),the mitochondria inner membrane anion carrier,may uncoupling the oxidative phosphorylation and ATP synthesis.To analyze the expression of UCP2 in human gastric adenocarcinoma tissues and investigate its correlation with the clinical pathological features and prognosis of gastric cancer patients,and to explore its effect on the proliferation and migration of gastric adenocarcinoma cells.MethodsTotally 142 cases of gastric cancer tissues and randomized 25 cases of adjacent normal tissues in Tianjin Medical University Cancer Institute and Hospital were included.We used immunohistochemical staining to detect the expression of UCP2 and to analyze its relationship with clinical pathological features.Kaplan-Meier method and Cox regression analysis were used to analysis the correlation between the expression of UCP2 and the outcome of gastric cancer patients.Western blot was used to detect UCP2 protein level after UCP2 siRNA transfection into SGC7901 gastric cancer cell lines.EdU assay and scratch assay were used to detect the proliferation and migration ability.Results1.The expression of UCP2 was significantly higher in gastric adenocarcinoma tissues compared with adjacent normal gastric mucosa tissues(65.5% vs.28%,P=0.006).2.The expression of UCP2 was significantly correlated with tumor infiltration(P=0.024),TNM stage(P=0.002)and lymph node metastasis(P=0.001),but had nothing to do with gender(P=0.954),age(P=0.362),size(P=0.756),position(P=0.964),and differentiation(P=0.633).3.The median survival of positive UCP2 staining is 42.9 months,compared with the 79.9 months of UCP2 negetive staining.The 5-year overall survival of patients with positive UCP2 expression was significantly lower than those without UCP2 expression(P=0.001).4.The proliferation and migration abilities were significantly inhibited with the knock down of UCP2 expression.Conclusion1.The significantly high expression of UCP2 in gastric adenocarcinoma tissues indicated that UCP2 played an important role in the gastric cancer progression and may be a prognostic factor of gastric adenocarcinoma patients.2.The silence of UCP2 expression inhibited the proliferation and migration of gastric cancer cells.UCP2 provided a new approach of gastric cancer target therapy.
