Objective: To our knowledge,the phospholipase C?1(PLCD1)is a potential tumour suppressor,which was studied in many tumours.And in pancreatic cancer cells,the cell cycle was inhibited and the apoptosis was promoted by PLCD1 expression plasmid.Now,to investigate the effects of PLCD1 on the proliferation,migration and invasion of pancreatic cancer cells.Methods: Liposome method was used to transfer the pc DNA3.1-PLCD1 to pancreatic cancer cells and the Vector group with empty vector was set.The proliferation?migratory and invasive abilities of pancreatic cancer cells were detected with CCK-8 assay?clone formation assay?transwell assay and wound healing assay.In additional,Western blot was used to demonstrate the potential mechanism.Results: Transient transfected with the PLCD1 over-expressing plasmid,PLCD1 was significant up-regulated in CAPAN-1 and BXPC-3 cells(P<0.05).The ability of proliferation showed significant inhibition with CCK8 and clone formation assay(P<0.05).The motility,invasion and migration were remarkable restrained with transwell assay and wound healing assay(P < 0.05).Additionally,Western blot showed PLCD1 induced the MAPK/ERK pathway molecule p-ERK1/2 downregulated(P<0.05),simultaneously,mesenchymal markers N-cadherin and vimentin was decreased(P<0.05).And increasing the expression of the epithelial marker E-cadherin(P<0.05).Conclusion: Our findings have presented the role of PLCD1 in pancreatic cancer,regulating the abilities of proliferation,migration and invasion,which may inhibit the MAPK/EKR pathway to suppress the EMT.This lay the foundation for the further study of PLCD1 in pancreatic cancer.
