| Small cell lung cancer(SCLC)is a highly malignant neuroendocrine tumor.The majority of SCLC cases are metastatic at the time of the first diagnosis,necessitating effective systemic therapies.While patients are sensitive to radiation and chemotherapy,they often relapse shortly after the cessation of treatment,the 5-year survival rate is extremely low.The second-line therapy of Topotecan response rate is limited and the duration of efficacy is short.There are no FDA-approved targeted therapies for SCLC,the progress in small-cell lung cancer has been hampered by the scarcity of specific molecular targets.Treatment and survival of patients with small-cell lung cancer has not changed substantially in more than 40 years.It reported that Notch signaling pathway plays a tumor suppressing role in SCLC,DLL3(delta-like protein 3)expressed in high-grade neuroendocrine lung tumors including SCLC is an inhibitory ligand of the Notch signaling pathway.DLL3 is a highly tumor-selective cell surface protein.Rovalpituzumab teserine(Rova-T)is a DLL3-targeted antibody-drug conjugate(ADC).Rova-T shows a significant and long-lasting effect in the treatment of patients with SCLC,especially high expression of DLL3. |
