Objective:To study the healing of pressure sore in rats with different levels of protein diet,and to further explore the role of protein in the healing process of pressure sore.Method:1.One hundred and forty healthy male SD rats were randomly divided into 5 groups?28 rats in each group?after they were purchased from the experimental animal center of hebei medical university.The rats were fed adaptively in the laboratory for one week?the temperature range of the laboratory was 20?-25??.The humidity ranges from 40 to 60.During adaptive feeding,rats were fed with standard rat food(the rat grain was purchased from the experimental animal center of hebei medical university,and the blood samples of each group of rats were collected from the tail vein of each group before the establishment of the model and recorded the contents of serum albumin and?Alb?in each rat.The difference was not statistically significant?P>0.05?,and the body weight of each rat was measured before the experiment.The difference was not statistically significant?P>0.05?.2.The rat model of pressure sore was made by referring to the method of Peirce,Wassermann et al.The method of placing aseptic iron tablets under the gluteal muscle of rats after 3 days of adaptation to sexual life in rats with sterile iron tablets and placing magnets under pressure in vitro[1-2],which forms a certain pressure between the magnets and the iron sheets.Apply pressure to the skin by pressing the tissue for 2hours to ischemia and hypoxia,then remove the magnet for 0.5 hours to allow tissue blood flow to be reperfused,5 cycles a day,then remove the magnet until the next day to continue with the above steps.After the establishment of the model,the rats in the five groups were given 10%crude protein and 20%crude protein respectively.3.The body weight,the area of pressure sore,the content of serum albumin and Alb in each group were measured one day,three days,seven days and fourteen days after the model.Finally,the healing time of pressure sore in each group was recorded.4.The statistical data were analyzed by spss19.0,and the meanąstandard deviation was used.The healing time was analyzed by single factor ANOVA,the area of pressure sore,body mass and serum Albumin werecomparedbyANOVA.Thecalibrationresultsof Greenhouse-Geisser were used.Result:1.30%protein group of rats in the intervention of high 30%protein diet during 7 days consecutive dead 12,the reason is due to anatomical analysis to find the rats eat high protein diet lead to indigestion,abdominal distension and intestinal obstruction and death due to residual content of the sample is too small,so removed 30%protein group.2.Comparison of the area of pressure sore:After injection of the 1days,3 days no significant pressure ulcer area of each rat size difference?P>0.05?.7 days and 14 days between the 10%protein group and 20%protein group,10%protein group and 25%protein group,15%protein group and 20%protein group,15%protein group and 25%protein group were statistically significant difference of pressure ulcer size?P<0.05?.The high protein pressure ulcer rats area is less than the low protein group rats.Four rats of pressure ulcer area of pressure ulcer area of longitudinal comparison,four groups of rats in the continuous decrease of pressure ulcer area.No significant differences between the 25%protein group and20%protein group?P>0.05?,but the pressure ulcer area than the 15%protein group and 10%protein group is smaller,the difference was statistically significant?P<0.05?.3.Comparison of body mass:Before moldle and after moldle of the 1day,there was no significant difference in body weight of rats in each group?P>0.05?.3 days,7 days,14 days,between 10%protein group and20%protein group,10%protein group and 25%protein group,15%protein group and 25%protein group was statistically significant difference in body weight of rats?P<0.05?,high protein group rats were higher than the weight of low protein rats.Body weight of rats in the four groups the longitudinal comparison,body weight of rats in the four groups have continued to increase,compared with four groups of rats body weight growth rate,25%protein group>20%protein group>15%protein group>10%protein group,10%protein group body weight growth the slowest,25%protein group body weight growth the fastest.4.Comparison of the content of serum albumin?Alb?:Each rat serum albumin?Alb?content into the mold before and first days after model had no significant difference?P>0.05?.3 days,7 days,the 14 day of 10%protein group and 20%protein group,10%protein group and 25%protein group,15%protein group and 25%protein group between the serum albumin?Alb?were statistically significant differences?P<0.05?,high protein group serum albumin?Alb?level is higher than the low protein group serum albumin?Alb?level.The four groups of rats serum albumin?Alb?each time the longitudinal comparison of 20%protein group and 25%protein group after model compared with serum Albumin?Alb?and before model content was no significant difference?P>0.05?.15%protein composition before model serum albumin?Alb?were compared with fourteenth days after model had significant difference?P<0.05?.10%protein composition before model serum albumin?Alb?and the content of After 3 days,7 days,14 days,the difference was statistically significant?P<0.05?.The intake of low protein diet in rats could lead to the continuous decrease of serum albumin?Alb?and prolong the healing time of pressure ulcer.5.Comparison of the healing time of pressure sore:20%protein group?d=16.36+1.57?<25%protein group?d=17.58+1.31?<15%protein group?19.27+1.27?<10%protein group?d=21.63+1.12?,healing time between the groups had significant difference?P<0.05?.Healing time of pressure ulcer in rats the slowest is 20%protein group,the longest is 10%protein group,one week before the incidence of pressure ulcer in rats is the fastest healing period.Conclusion:1.The supply of protein in the healing process of pressure sore is not the more the better,but the best protein intake.2.The best quality of protein supplemented in the fastest period of granulus growth in the early stage of pressure ulcers is best for the healing of pressure sores.3.There is no correlation between the mass of body and the healing time of pressure sore.4.The change in the content of Abl can be used as a monitoring indicator for predicting the prognosis of pressure sore.5.the long-term effect of nutritional intervention on pressure sore is obvious.
