Objective To have a knowledge of the expression discrepancy of Lnc RNA-00704 between cc RCC and para-tissues;to find out the relationship between Lnc RNA-00704 expression and cc RCC patients prognosis;to study how it affects the proliferation,transmission and metastasis of cc RCC cell lines.Methods We firstly search the TCGA data base for the data of renal carcinomas,compare the expression level of Lnc RNA-00704 in cc RCC with that in para-tissues and find out the expression of Lnc RNA-00704 is associated with cc RCC clinic pathological parameters.to analyze the relationship between the expression of Lnc RNA-00704 and patient' survival with the Log-rank test and illustrated it with the Kaplan-meier survival curve.Secondly,we test the expression of Lnc RNA-00704 in 45 pairs of clinical specimens,in four cc RCC cell lines(786-O,ACHN,OS-RC-2,CAKi-1)and in nomal renal cell HK2,then the outcomes are used to testify the outcomes of online analysis.Lastly,we construct expression vector of sh RNA to investigate the effect of Lnc RNA-00704 expression on the biological property of cc RCC cell lines,including their proliferation,migration and metastasis.Outcomes 1.The TCGA data base search show the expression of Lnc RNA-00704 in cc RCC is relatively higher(2.4times,P<0.01),its expression is not correlated with gender(p=0.115),age(p=0.108),recurrence(p=0.009);but it has notable correlations with cc RCC pathological grades (p=0.015),TNM stage(p=0.001)and metastasis(p=0.009).the patients with high Lnc RNA-00704 expression have a notably worse prognosis than those with low Lnc RNA-00704 expression.2.Cell biological function test show the proliferation,migration and invasiveness of cc RCC is suppressed after Lnc RNA-00704 genetic expression is interrupted.Conclusions 1.Lnc RNA-00704 has a higher expression in cc RCC and has a notable correlation with cc RCC pathological grades,TNM stage and metastasis,it suggests Lnc RNA-00704 may play an role in the occurrence and advance of cc RCC,therefore,to test its expression help to diagnosis this disease in early stage and to predict its prognosis.2.The proliferation,migration and invasiveness of two cc RCC cell lines(ACHN,786-O)is suppressed after Lnc RNA-00704 genetic knockdown,it suggests Lnc RNA-00704 which may act as a role of oncogene has important biological functions,thus we may get a clue to explore new way to treat this disease.
