Objective:To study the therapeutic effect of moxibustion(Mox)on ulcerative colitis(UC)mice and the oral caffeine influence on moxibustion induced anti-inflammatory effects of UC.Methods:All mice 32±2g were randomly divided into control group,model group(DSS group),moxibustion group,caffeine group and caffeine+moxibustion group.Mice in model group,moxibustion group,caffeine group and caffeine+moxibustion group were induced with 3% Dextran Sulfate Sodium(DSS)-mediated ulcerative colitis.Mice were treated with moxibustion therapy or caffeine(0,10,30,50mg/kg/day),subsequently analyzed colitis development.Statistical Analysis: Weighted data was analyzed by repeated measures analysis of variance,and the DAI score was analyzed by one-way analysis of variance.P<0.05 was considered statistically significant.Results:Mice treated with moxibution displayed benefit response towards DSS-induced colitis,characterized by lower body weight loss,disease activity index(DAI)scores and histological manifestations.Caffeine resulted in the reduction of UC in a caffeine no-dose dependent manner.However,50mg/kg/day caffeine inhibits moxibustion anti-inflammation effects on UC mice.1.Efficacy evaluation of moxibustion on UC mice:Mice treated with moxibution displayed benefit response towards DSS-induced colitis,characterized by lower body weight loss,disease activity index(DAI)scores and histological manifestations.2.Efficacy evaluation of UC mice treated with different doses of caffeine:Caffeine resulted in the reduction of UC,characterized by lower DAI scores and histological manifestations.However,caffeine doses(10,30,50mg/kg/day)showed similar efficacy in UC mice(P>0.05).3.Effect of caffeine on moxibustion treatment of UC mice:(1)There was no significant difference in DAI score between 10mg/kg/day caffeine+moxibustion and 30mg/kg/day caffeine+moxibustion group(P>0.05).The DAI score was significantly higher in 30mg/kg/day caffeine+moxibustion group than in the moxibustion group(P<0.05).(2)The results of light microscopy showed that mucosal edema was rarely seen in the low concentration + moxibustion group,moderate concentration + moxibustion group,a small amount of glandular destruction,a small amount of inflammatory cell infiltration;high concentration + moxibustion group due to the concentration,high concentration due to submucosa rare Congestion or edema,glands arranged more neatly,a small amount of inflammatory cell infiltration.Conclusions:1.Moxibustion has a therapeutic effect on UC.2.Development of acute ulceative colitis is reduced upon caffeine treatment,no matter the different caffeine intake dose.3.High doses of caffeine intake can inhibit the efficacy of moxibustion,moxibustion treatment of UC should try to avoid high doses of caffeine intake.
