| Objective: This study aims to analyze the expression of SMYD2 in lung cancer cells and lung cancer tissue,and to discuss their relationships with clinicopathological features and prognosis.We further explored the relationship between SMYD2 and post transcriptional modification of p53 and PTEN,and the relationship between p53K370 mel and PTENSer380 pi and survival prognosis of patients with lung cancer.Methods:Paraffin embedded 101 lung cancer tissue and 41 normal lung tissue were collected and made into tissue chips.The expression of SMYD2 in lung cancer cell lines was detected by immunohistochemistry.The expression of p53 and PTEN was detected by Western blotting and immunohistochemistry.And we definited the optimal concentration of SMYD2,p53,p53K370 mel,PTEN and PTENSer380 pi protein expression in tumor tissues.The expression of SMYD2,p53,p53K370 mel and SMYD2,PTEN,and PTENSer380 pi were detected by multiplex staining on two tissue microarrays.The correlation of SMYD2,p53,p53K370 mel,PTEN and PTENSE 380 pi proteins were analyzed.The correlation of Sp53K370 mel,PTENSer380pi and the prognosis of OS and DFS in patients with lung cancer was analyzed.Results: 1.The expression of SMYD2,p53 and PTEN in lung cancer tissue and normal lung tissue and their correlation with clinicopathological characteristics:Compared with normal lung epithelial cells,SMYD2 showed high expression,while p53 and PTEN showed decreased expression.SMYD2 were mainly expressed in the cytoplasm and nucleus of lung cancer cells.There were significant higher in the expression of the SMYD2 in the tumor tissue compared with the adjacent tissue(P=0.003).The expression of the p53 and PTEN was significant higher in the paracancerous tissues than in the lung cancer tissues(P<0.001 and P=0.010).No significant differences were observed between the expression of SMYD2 and the patient age,smoking status,tumor location,pathological type,TNM stage as well as lymph node metastasis status(P>0.05).2.To analyze the relationship between SMYD2 and p53,PTEN and their post-transcriptional modification on twomulti-stained tissue chips: We analysis the correlation among the expression of SMYD2,p53,and p53K370me1 in lung cancer tissues on a tissue chip.From the analysis results,there was a significant negative relevance between SMYD2 and p53 in lung cancer tissues(rs=-0.259,P=0.029).Meanwhile,p53 and p53K370me1 showed a significant negative relevance in tumor tissues(rs=-0.508,P=0.000,P<0.01).In lung cancer tissues,there was a significant positive relevance between SMYD2 and p53K370me1(rs = 0.531,P = 0.000,P <0.01).We also analysis the correlation among the expression of SMYD2,PTEN,and PTENSer380 pi in lung cancer tissues on another tissue chip.As shown in the results,there was a significant negative correlation between SMYD2 and PTEN in lung cancer tissues(rs=-0.306,P=0.007).Meanwhile,the PTEN and PTENSer380 pi proteins showed a significant negative correlation in tumor tissues(rs=-0.056,P=0.032).There was a significant positive correlation between SMYD2 and PTENSer380 pi in lung cancer tissues(rs=0.343,P=0.002).3.The survival prognostic analysis of SMYD2,p53k370me1 and PTENser380 pi in patients with lung cancer in DFS and OS: the prognosis of the high-expression group of SMYD2,p53K370me1,and PTENSE380 pi was poor compared with the low expression group,and the difference was statistically significant.Conlusion: SMYD2 show high expression in lung cancer cell lines and lung cancer tissues.SMYD2 is associated with poor prognosis.2.There was a significant negative correlation between SMd p53K370pi;p53 and p53K370 pi showed significant negative correlation.3.There was a significant negative correlation between SMYD2 and PTEN in lung cancer tissues;SMYD2 was positively correlated with PTENSer380pi;PTEN and PTENSer380 pi showed a significant negative correlation. |
