Objective: It was to study the expression of ribosomal protein L29 in gingival squamous cell carcinoma(GSCC)and investigate clinical pathology significance of RPL29 in GSCC.Methods: A total of 50 cases of gingival cancer were collected from Oral and Maxillofacial Surgery,Affiliated Hospital of Qingdao University.Tumor tissues,para-cancer tissues, and normal gingival tissues were collected.Twenty-one cases of tongue cancer tissues were used to detect the expression of RPL29 protein by SP immunohistochemical method.RPL29 was analyzed. The clinical pathological significance of gingival cancer was explored in different clinical T stages, pathological grades, lymph node metastases, and gender, age, smoking history, drinking history.Result: The positive expression rate of RPL29 in gingival cancer was 66.67%,which was significantly higher than that of adjacent tissue(19.05%),and it was not expressed in normal gingival tissue(positive rate was 0%).The expression of RPL29 was significantly different(P<0.05).There was no statistically significant difference between the positive expression of RPL29 and the different clinical T stages and pathological grades of gingival cancer. The positive rates of RPL29 in T1,T2,and T3+T4 were 81.82%(9/11),86.67%(26/30),and 66.67%(6/9),respectively,with no statistically significant differences(P>0.05).The positive rates of RPL29 in highly differentiated gingival cancer(36 cases)and lower differentiated gingival cancer(14 cases)were 77.78%(28/36)and 92.86%(13/14),respectively,with no statistically significant difference(P>0.05).The positive rate of RPL29 was 100%(15/15)in patients with lymph node metastases and 74.29%(26/35)in those without lymph node metastases. There was a significant difference in the expression of RPL29 between patients with and without lymph node metastases in gingival cancer patients.Significance(P<0.05).The expression of RPL29 was not significantly different in independent groups(P>0.05),such as different age groups, gender, smoking, and drinking history. The positive expression rate of RPL29 in tongue cancer was 85.71%,which was significantly higher than that in paratumor tissue. There was a significant difference in the expression of RPL29 between lymph node metastasis group and tongue cancer group(P<0.05),This expression is basically similar to that of gingival cancer.Conclusion:(1)RPL29 was highly expressed in gingival squamous cell carcinoma. It was lowly expressed in paracancerous tissues and not expressed in normal gingival tissues, suggesting that RPL29 may be a promoter of gingival squamous cell carcinoma and can be used as a molecular biomarker for the detection of gingival squamous cell carcinoma.(2)The expression of RPL29 was positive in gingival squamous cell carcinoma with cervical lymph node metastasis. The squamous cell carcinoma with negative RPL29 expression had no lymph node metastasis.RPL29 was closely related to the occurrence of cervical lymph node metastasis in gingival squamous cell carcinoma.(3)The expression of RPL29 in gingival squamous cell carcinoma was not related to age, sex, smoking history, drinking history, tumor site, clinical T stage, and case grading.(4)RPL29 is highly expressed in tongue squamous cell carcinoma and low in paracancerous tissues. It is closely related to cervical lymph node metastasis in tongue squamous cell carcinoma and similar to gingival squamous cell carcinoma.RPL29 was closely related to oral cancer.
