ObjectiveAccording to researches,FBP1 has been found to play a key role in the development and progression of tumor in several types of cancers,including lung cancer,breast cancer,renal carcinoma and gastric cancers.According to our previous research,we screened the differentially expressed proteins in three hepatocellular carcinoma cell lines with different metastatic potential,and glycolysis/gluconeogenesis ranked at the top position.Thus,we hypothesize that FBP1 affects invasion of hepatocellular carcinoma cells.Therefore,this study aims to investigate effects of FBP1 on the biological characteristics of MHCC97-H hepatocellular carcinoma cells by knocking down FBP1 in MHCC97-H cell.We attend to explore whether FBP1 is a targeted gene that affects the biological characteristics of hepatoma cells and provides experiments for the molecular targets of inhibiting liver cancer metastasis and invasion.Methods1.Verify the differential expression of FBP1 between MHCC97-H,HCCLM3 and MHCC97-L cell lines;2.Lentivirus infected hepatocellular carcinoma cell MHCC97-H,and screened and verified stabled FBP1 i knockdown cells;3.In vivo and in vitro tumor biological behavior of FBP1 i cell line was verified.Results1.FBP1 expression detected by WB assay in MHCC97-H cell line was higher than HCCLM3 and lower than MHCC97-L cell lines;FBP1 protein of stabled FBP1 knockdown cells was significantly reduced compared with control groups;2.There was no significant difference in cell growth,cycle,apoptosis and migration between FBP1 i and control groups(p>0.05);The production of lactic acid in FBP1 i was significantly higher than that in control groups(p.<0.05),Mitochondrial aerobic respiration was decreased compared with control groups;3.No metastasis was found in nude mice liver inoculation experiments.ConclusionsFBP1 had no significant effects on the invasion and migration of hepatoma MHCC97-H cells.
