Objective:To investigate the effect of human hepatic stellate cells on the cytotoxicity of ?? T cells from healthy volunteers and hepatocellular carcinoma patients,and the effect of ?? T cells on the proliferation,invasion and migration of hepatocellular carcinoma.Methods: Density gradient centrifugation and flow cytometry were used to obtain ?? T cells;phytohemagglutinin and interleukin-2 were used to stimulate the proliferation of ?? T cells;human hepatic stellate cells and ?? T cells were co-cultured with transwell chamber;ELISA was used to detect the changes of cytokine INF-?;CCK-8 assay was used to detect the proliferation of HepG2 cells;Transwell chamber assay was used to detect the invasion and migration ability of HepG2 cells.Results: After co-culture with HSC,the cytotoxicity of ?? T cells from healthy volunteers and patients with HCC was enhanced,and the former is stronger than the latter(P<0.05);IFN-? secreted by ?? T cells was increased,and healthy volunteers group secreted more than those with HCC(P<0.05);The ability of ?? T cells to inhibited the proliferation,invasion and migration of HepG2 cells was enhanced,and the healthy volunteers were stronger than those of the patients with HCC(P<0.05).Conclusion: HSC can promote ?? T cells' secretion of IFN-?,enhance the cytotoxicity of ?? T cells and the ability of ?? T cells to inhibit the proliferation,invasion and migration of HCC,and the enhancement effect is related to the source of ?? T cells. |
