The principle of photodynamic therapy(PDT)is caused by reactive oxygen species(ROS),which is activated by specificity concentrated of photosensitizer in tumor tissues after a specific wavelength light irradiate,then through the oxidative damage created by reactive oxygen species.The effects of the oxidative damage by Reactive oxygen species destroy structure and function of organelles at target sites,It can cause apoptosis and necrosis of target cells to play a therapeutic role.Because of its small trauma,no limitation of the number of skin lesions,repeatable treatment and so on,it has been widely used in superficial basal cell carcinoma of the skin,actinic keratoma and other skin diseases treatment is a more promising treatment of dermatology.However,the efficacy of PDT alone in the treatment of skin diseases such as nodular basal cell carcinoma is limited.It is not only affected by the penetration depth of photosensitizer and the depth of red light penetration,but also may be related to the PDT resistance of tumor cells.At present,Nuclear factor erythroid2-related factor 2(Nrf2)and TFIIIB-related factor 2(Brf2)have been studied more frequently.The Nrf2 is a key factor in oxidative stress response.It plays an important role in antioxidant damage of many cells,tissues and organs by combining downstream antioxidant elements.TFIIIB related factor 2 is a gene encoding BRF2 protein.Currently,it has been found that BRF2 can directly participate in oxidative stress stimulation pathway,and overexpression can induce acquired ROS resistance in tumor cells,which plays a key role in the defense system induced by oxidative stress.It may be involved in the production of PDT resistance in tumor cells during PDT therapy.This article will summarize the relationship and research progress of Brf2 gene,Nrf2 signaling pathway and photodynamic therapy in Department of dermatology.
