Objective:Microplasma radiofrequency technique(Plasma)has been successfully applied to scar,skin,wrinkle,photoaging and so on,and intense pulse light pulsed(IPL)has been widely used in the treatment of skin pigmentation,wrinkle,and so on.However,there are few reports on the treatment of skin pigmentation by microplasma radiofrequency technique(Plasma).In this study,we compared the therapeutic effects of microplasma radiofrequency(Plasma)and intense pulsed light(IPL)on brown skin pigmentation induced by NB-UVB in the back of pigmented guinea pigs.The therapeutic effect was evaluated by observing the changes of melanin granules and?-MSH and MC1R in epidermis to provide the experimental theoretical basis for the treatment of skin pigmentation with Plasma.To provide more options for the future treatment of skin pigmentation disease.Methods:10 healthy floral guinea pigs(clean grade,weight 200~250g)were shaved and exposed to skin in the brown area of the back of guinea pigs.Each guinea pig was irradiated with narrow band ultravioletine NB-UVB at a dose of 900mJ/cm~2per week for 3 weeks.The total dose of naked skin irradiation on the back of each guinea pig was 2700mJ/cm~2.After the establishment of the model,all the molded areas of the back were randomly divided into three areas with the shape of"Pin",which were treated differently,with the head of each guinea pig facing toward the top:1.The lower left region was blank control group A,and no treatment was done after the model was formed.2.The above area was treated with IPL group B,and the model area on the back of guinea pig was irradiated by IPL.The treatment parameters were as follows:wavelength 570-950,spot area 10mm×30mm,pulse width 12ms,energy density 8mJ/cm ~2;3.The lower right region is Plasma treatment group C,the exposed skin tissue in this area is irradiated with the fixed point treatment head.The treatment parameters are as follows:the output power is 30Watt,the time of action is 0.6s,the output energy is 18J,respectively,before modeling,The plasma and IPL group and the blank control group were treated with IPL immediately.After 1 week,2 weeks,3 weeks and 4 weeks after treatment,the changes of pigmentation were assessed and pathological sections were made.Melanin granules were stained with Masson-Fontana and?-MSH and MC1R were stained with immunohistochemistry.The changes of the above indexes and the changes of different indexes at different time points in the same group were observed to explain the therapeutic effect of each treatment method.Results:General observation and evaluation:After NB-UVB irradiation,the bare areas in the back of guinea pigs showed a dark brown and black color with uniform distribution of pigments.The changes of pigmentation were not obvious at the first week after treatment,at the second week after treatment,and at the third week after treatment.The pigmentation gradually disappeared at the 4th week after treatment,especially in group B and C,especially in group B,which was more obvious than that in group A,and the fading of group B was more obvious than that of group B.Changes of melanin granules:Compared with normal skin tissue before and after NB-UVB irradiation,the content of melanin granules increased significantly(P<0.001).Immediately after treatment,1 week after treatment,2 weeks after treatment,3weeks after treatment.At 4 weeks after treatment,the content of melanin granules in group B and group C was lower than that in group A,and the difference was statistically significant(P<0.001)and the content of melanin granules in group B was lower than that in group C.The difference was statistically significant(P<0.001).There was significant difference in the content of melanin particles between groups(P<0.001)and within groups(P<0.001).Changes of?–MSH,MC1R:The changes of?-MSH and MC1R were detected by immuno-histochemical staining.The results showed that the expression of?-MSHU MC1R in skin tissue increased after irradiation with NB-UVB,which was significantly different from that in normal skin tissue before irradiation(P<0.001).1.?-MSH:immediately after treatment,1 week after treatment,2 weeks after treatment,3 weeks after treatment and 4 weeks after treatment,the expressionS of?-MSH in groupB and C were lower than that in group A.The difference was statistically significant(P<0.001),and the expression of group B was lower than that of group C(P<0.001).2.MC1R in group C was lower than that in group A immediately after treatment,1 week after treatment,2 weeks after treatment,3 weeks after treatment and 4 weeks after treatment.The difference was statistically significant(P<0.001),and the expression of group B was lower than that of group C(P<0.001).Conclusions:1.The Intense pulsed light and the microplasma radiofrequency technique can reduce the pigmentation in the dorsal skin of guinea pigs induced by NB-UVB.2.Intense pulsed light and microplasma radiofrequency techniques can reduce the expression of?-MSH MC1R in dorsal skin pigmentation induced by NB-UVB in guinea pigs.3.The microplasma radiofrequency technique and intense pulsed light therapy can promote the skin pigmentation of guinea pigs induced by NB-UVB,but the microplasma laser has a weaker effect on this aspect.
