Background and purpose:CIN85 is a member of the signal adapter protein family,which is extensively involved in various physiological and pathological activities of cells and is involved in the development and progression of tumors.Up to now,abnormal expression of CIN85 was detected in many cancers such as colon cancer,breast cancer,oral squamous cell carcinoma,glioma and melanoma,and it was found to be associated with the growth,migration and invasion of various cancer cells,but no report in esophageal cancer.The purpose of this study is: 1.To investigate the expression of signal adapter protein CIN85 in esophageal squamous cell carcinoma and normal esophageal tissues and to analyze the related clinicopathological factors.2.To study the effect of CIN85 on the cytology of esophageal squamous cell carcinoma.Methods: 1.Immunohistochemistry was used to detect the differential expression of CIN85 in 43 cases of esophageal squamous cell carcinoma and adjacent normal tissues.2.The expression of CIN85 in esophageal squamous carcinoma cell line TE1 was reduced by sh RNA interference technology.MTS assay,wound healing assay,Clone formation assay and transwell assay were used to detect the influence of CIN85 expression on tumor cytology,such as proliferation,migration and invasion of esophageal cancer cells.Results: 1.The expression of CIN85 in esophageal squamous cell carcinoma was significantly higher than that in adjacent normal tissues,and there was a significant correlation between the expression of CIN85 and the presence of lymph node metastasis and late staging.2.After sh RNA interference technology to reduce the expression level of CIN85,MTS assay showed that the decreased expression of CIN85 significantly inhibited the proliferation of esophageal cancer cells;The wound healing assay showed that the expression of CIN85 significantly inhibited the migration of esophageal cancer cells;The clonal formation ability of esophageal cancer cells was significantly decreased;Transwell experiments showed that the expression of CIN85 decreased esophageal cancer cell invasion decreased significantly.Conclusion: In conclusion,the present study shows that the expression of the signal adapter protein CIN85 is abnormal in esophageal squamous cell carcinoma and has significant correlation with lymph node metastasis and tumor TNM staging.CIN85 is involved in the development of esophageal squamous cell carcinoma.Inhibition of CIN85 expression,can inhibit the proliferation,migration and invasion of esophageal cancer cells.CIN85 may be a potential molecular target for the treatment of esophageal cancer.
