Objective: With the progress of society,the development of industry and because of people's bad living habits,lung cancer has become one of the diseases that seriously threaten human health.The latest statistics released by the National Cancer Center shows that lung cancer has become the top cancer that morbidity of malignancy and death of malignant tumors nationwide.Therefore,it is of great significance to develop research of the lung cancer related genes and the mechanism of incidence and metastasis of lung cancer in order to further improve the diagnosis and treatment of lung cancer.The Sparcl1 gene,also known as MAST9 or hevin,is a member of the Sparc family and also an extracellular matrix.With the size of 47 KB,the gene encodes a protein with a molecular weight of about 71 KDa.Sparcl1 genes were expressed in most organizations,with higher amount of expression especially in lung tissue,oarium,skeletal muscle and cerebrum,relatively lower amount of expression in Placenta,pancreas and thymus and no expression is relatively low in hepar and Peripheral blood leukocyte.Therefore,Sparcl1 gene plays an important role in human development and organ formation.The Sparcl1 gene,also known as MAST9 or hevin,is a member of the Sparc family and also an extracellular matrix extracellular matrix.With the size of 47 KB,the gene encodes a protein with a molecular weight of about 71 KDa.Sparcl1 genes were expressed in most organizations,with higher amount of expression especially in lung tissue,oarium,skeletal muscle and cerebrum,relatively lower amount of expression in Placenta,pancreas and thymus and no expression is relatively low in hepar and Peripheral blood leukocyte.Therefore,Sparcl1 gene plays an important role in human development and organ formation.Recent years it has been found in the researches home and abroad that the Sparcl1 genes inhibits Tumor invasion,migration and proliferation of the Tumor and are also involved in blood vessel formation,cell shaping and cell adhesion.Researchers home and abroad found that the gene is in relatively low expression in most tumor tissues,but in increasing expression in glioma.At home and abroad has not yet test Spaecl1 gene expression levels of lung cancer cells and not reported the role in which it is in the cell cycle,and did not validate Sparcl1 expression in lung tissue of non-small cell lung cancer and its clinical significance.Quantitative real-time PCR,western-blot are used as well as immunohistochemistry detecting the expression of Sparcl1 non-small cell lung cancer tissues,which is to analyze its clinical significance.The biological function of the gene is detected through transferring overexpression of Sparcl1 Gene plasmids into A549 cells in lung adenocarcinomaMethods: 1.Quantitative real-time PCR and Western-blot is used to detect the expression of Sparcl1 gene in non-small cell lung cancer and verify the process of transfection.2.Immunohistochemistry is used to detect the expression of Sparcl1 gene in non-small cell lung cancer tissues.3.Cell Scratch Test and flow cytometer(FCM)are used to detect the influence on lung adenocarcinoma A549 cells' migration,apoptosis and cell cycle causing by the overexpression of lung adenocarcinoma A549 Cell and negative control group.Results: 1.The expression of Sparcl1 m RNA in non-small cell lung cancer tissues is lower than that in lung tissue of para-cancer.2.The expression of Sparcl1 protein in non-small cell lung cancer tissues is relatively lower than that of the adjacent lung tissue.3.The migration ability of overexpressed Sparcl1 lung adenocarcinoma cells is weaker than that of the negative control group.4.The number of cells overexpressed of apoptosis of Sparcl1 lung adenocarcinoma is higher than that of the negative control group.5.Overexpressed Sparcl1 lung adenocarcinoma cells in G1 stage is higher than that of the negative control group,but G2 and S stage is lower than that of the negative control group.6.The expression differences of Sparcl1 in non-small cell lung cancer are related to clinical stages.Patients with low expression of Sparcl1 had a 5-year survival rate lower than those with high expressionConclusion: 1.Sparcl1 gene is in low expression in non-small cell lung cancer tissues.The lower expression of this gene indicates the higher clinical staging,and the lower survival rate in five years.2.Changes in the expression of Sparcl1 gene affect the ability of cell migration,apoptosis and cell cycle in lung adenocarcinoma A549 cells.
