Objective: Effect of different dosage of tea polyphenol on the expression of TNF-α,IL-1β,IL-6,IL-8 in serum and NF-κB in gingival tissue of diabetic periodontitis rats has been investigated in the present work,aiming at investigating the effect of tea polyphenols on diabetic periodontitis and providing a theoretical reference for tea polyphenols as an adjuvant therapy drugs of diabetic periodontitis.Methods:1.Eighty male rats weighted ranging from 140 g to 160 g are applied to divide into 4groups randomly: blank group(10 rats),diabetes group(10 rats),periodontitis group(10rats)and model group(50 rats).2.Periodontitis model was established by periodontal ligation + high sugar soft diet feeding.Type 2 diabetes model was established by feeding with high fat and high sugar diet for 5 weeks + intraperitoneal injection of STZ(30 mg/Kg).Type 2 diabetic periodontitis model was established by periodontal ligation + feeding with high fat and high sugar diet for 5 weeks + intraperitoneal injection of STZ(30 mg/Kg).3.Thirty-six rats were established the diabetes periodontitis model successfully.Rats were divided into 4 groups randomly: diabetic periodontitis group,low dose group,middle dose group and high dose group,and each group consists of 9 rats.Blank group,diabetes group,periodontitis group and diabetic periodontitis group were fed gavage daily with 10 m L/kg double distilled water,low dose group was fed gavage daily with 400 mg/(kg·d)/10 m L TP,middle dose group was fed gavage daily with 800 mg/(kg·d)/10 m L TP,and high dose group was fed gavage daily with 1600 mg/(kg·d)/10 m L TP.4.Four weeks of intragastric administration,tail tip were cut to collect blood sampling after fasted 12 h,and collect blood serum after centrifugation.Eight weeks of intragastric administration,all rats had a intraperitoneal injection anesthesia after fasted 12 h,then collected blood from the ventral main vein,and rapid separation of the gingival tissue from bilateral maxillary second molar.TNF-a,IL-1β,IL-6,IL-8 and Hb A1 c in rats serum has been investigated by ELISA.The expression of NF-kappa B in gingival tissue of rats has been investigated by Western Blot.Results:1.Establishment of animal modelPeriodontitis group and model group,the alveolar bone in the second molar area was absorbed and the periodontal pocket formed,and the periodontal destruction of model group was more serious.All showed that the periodontitis model was established successfully.Diabetes group and model group,fasting blood glucose of rat was more than11.1 mmol/L.All showed that the diabetes model was established successfully.2.Detection of rat serumThe content of TNF-αin diabetes group and periodontitis group was significantly higher than that in blank group(P<0.05).Diabetic periodontitis group was significantly higher than that in diabetes group and periodontitis group(P<0.01);Low dose group and middle dose group were lower than diabetic periodontitis group(P<0.05)(P<0.01),and the decrease of middle dose group was more obvious(P<0.05).The level of IL-8 in diabetes group and periodontitis group was significantly higher than blank group(P<0.05).Diabetic periodontitis group increased further(P<0.01);After the TP intervention,the level of IL-8 decreased in varying degrees(P<0.01),and there was no statistical difference between middle dose group and blank group(P>0.05).The content of IL-6 in periodontitis group and diabetes group was higher than that in blank group(P<0.05)(P<0.01);diabetic periodontitis group increased further(P<0.01).Compared with the diabetic periodontitis group,all TP intervention groups were decreased(P<0.01),and the most significant reduction was in the middle dose group(P<0.01),and close to the blank group(P>0.05).The content of IL-1 β was no statistical difference between diabetes group and blank group(P>0.05);the content of IL-1βin periodontitis group was higher than blank group(P<0.05),and diabetic periodontitis group increased further(P<0.01).After intragastric administration,low dose group and middle dose group were decreased(P<0.01),and there was no statistical difference between low dose group and middle dose group(P>0.05).The content of HbA1 c in diabetes group and diabetic periodontitis group was higher than blank group(P<0.05)(P < 0.01),there was no statistical difference between periodontitis group and blank group(P>0.05).After the TP intervention,the content of Hb A1 c in low dose group and middle dose group were decreased(P<0.01),and the decrease of middle dose group was more obvious(P<0.01).3.Expression of NF-κB in gingival tissue of ratsThe expression of NF-κB in periodontitis group was significantly higher than blank group(P<0.05).Diabetes group was higher than blank group,but the difference was not statistically significant(P>0.05).Low dose group and middle dose group were lower than diabetic periodontitis group(P<0.01).There was no statistical difference between the high dose group and the diabetic periodontitis group(P>0.05).Conclusion:1.Tea polyphenols can reduce the level of HbA1 c in serum of diabetic periodontitis rats,and it has a certain hypoglycemic effect.2.Tea polyphenols can inhibit the expression of NF-κB in gingival tissue of diabetic periodontitis rats,and reduce the levels of TNF-α,IL-1β,IL-6 and IL-8 in serum of rats.
