ObjectiveTo investigate the changes of behavior,oedema condition of brain tissue,the change of intestinal function,and the protein change of vascular endothelial growth factor?VEGF?and cluster of differentiation 34?CD34?in aged rats with traumatic brain injury,and to provide the reference for research and treatment of aged patients with traumatic brain injury.MethodsRat models with traumatic brain injury were made by the method of Feeney.All rat models with traumatic brain injury were divided into 5 groups according to injury time,and they were respectively 3-hours Group,12-hours Group,24-hours Group,72-hours Group and 5-days Group with 10 rats in each group.Ten rats with sham surgery in which brains were not stroked by freely falling objects after incision of scalp were selected as Control Group.The behaviors in all groups were evaluated by Nerological Severiyt Score?NSS?.The brain water content were determined by dry-wet method.The mucous membrane of small intestine in all groups were observed after instenal section were made and dyed by HE staining.The indictors of jejunum barrier function?the villi height,villi diameter,crypt depth and the villi erea of jejunum?in all groups were observed under the light microscope.The protein expression of VEGF and CD34 of brain tissue in all groups were observed after brian section were made and dyed by immumohistochemical staining.Results?1?Compared with Control Group,the score of NSS was significantly increased in 5traumatic brain injury groups?P<0.05?,and the score of NSS reached the peak in 72ndd hours and 5th day after brain injury?P<0.05?.?2?Compared with Control Group,the brain water content was significantly increased in 5 traumatic brain injury groups?P<0.05?,and the brain water content reached the peak in 24th,72ndd hours and 5th day after brain injury?P<0.05?.?3?Compared with Control Group,the destruction of the intestinal mucosa villi,the loose villous stroma,intestinal villi regularly arranged even rupture,villus epithelial necrosis,shorter,spacing widened,lamina propria edema occured,and all above abnormality reached the peak in 24th and 72ndd hours day after brain injury?P<0.05?.?4?Compared with Control Group,the villi height,villi diameter,crypt depth and the villi erea of jejunum was significantly decreased in 5traumatic brain injury groups?P<0.05?,and the brain water content reached the valley in 72ndd hours and 5th day after brain injury?P<0.05?.?5?Compared with Control Group,the protein expression of VEGF and CD34 in brain tissue was significantly increased in 5 traumatic brain injury groups?P<0.05?,and the brain water content reached the peak in 72ndd hours and 5th day after brain injury?P<0.05?.ConclusionWith the extension of injury time,aged rats with traumatic brain injury,increased behavioral score and water content in brain tissue have been along with increased VEGF and CD34 protein expression in brain tissue.From 24 hours to 72 hours after injury,injury reached a peak.Reducing protein expression of CD34 and VEGF in brain tissue may help improve symptoms of traumatic brain injury,which might provide a new target for clinical treatment.
