Objective: To investigate the molecular mechanism of the development of epithelial ovarian cancer(EOC),the expression profiles and functions of Long non-coding RNAs(LncRNAs)and mRNAs in ovarian epithelial cancer cell lines were analyzed.Methods: First,the differentially expressed LncRNAs and mRNAs were scanned by LncRNA gene microarray among the epithelial ovarian cancer cell lines A2780,HO8910,SKOV3 and ovarian epithelial cell line HOSEpiC.Meanwhile,the altered genes in the three epithelial ovarian cancer cell lines were analyzed by Gene Ontology(GO)and KEGG enrichment pathway.Subsequently,the dominant differently expression of LncRNAs were verified by qRT-PCR among the epithelial ovarian cancer cell lines A2780,HO8910,SKOV3,OVCAR3,3AO cell lines and ovarian epithelial cell line HOSEpiC.Then,the expression of LncRNAs RNASEH1-AS1 s and KB-1836B5.1 were verified by qRT-PCR between the epithelial ovarian cancer and ovarian tissues.At the end,the effect of knock-down RNASEH1-AS1 and over-expression KB-1836B5.1 in ovarian cancer cell lines,were verified by Edu cell proliferation examination and transwell assays.Significant enrichment signaling pathway was detected by western blot.Result: 227 LncRNAs were up-regulated and 483 LncRNAs were down-regulated,and 359 mRNAs were up-regulated and 148 mRNAs were down-regulated in three cancer cell lines respectively.GO analysis indicated the molecular function of differential genes mainly involved in signal transduction and protein kinase activity.The KEGG pathway analysis revealed that the altered genes in the three epithelial ovarian cancer cell lines were mainly enriched in PI3K-AKT,mTOR and TNF? pathways.The different expression of LncRNAs were selected and evaluated by RT-PCR.The results were consistent with that of gene microarray.RNASEH1-AS1 was up-regulated,but KB-1836B5.1 was down-regulated in epithelial ovarian cancer.Knock-down RNASEH1-AS1 inhibits epithelial ovarian cancer cells proliferation,migration and invasion.The expression of p-AKT?p-mTOR and p-p70S6 K were decreased in SKOV3 with RNASEH1-AS1 Knock-down.The same results were found in HO8910 with KB-1836B5.1 over-expression.Conclusion:It was inferred that RNASEH1-AS1 and KB-1836B5.1 were involved in the oncogenesis and differentiation.
