Objective:Preliminary exploration of LINC00473(long noncoding RNA00473,LINC00473)in gastric cancer cell lines,normal gastric mucosa cells,normal gastric tissues and gastric cancer tissues.The migration,and proliferation ability and cell apoptosis of gastric cancer cells for clinical provides the basis for the treatment of gastric cancer patients.Methods: Using the method of q RT-PCR to detect LINC00473 expression in normal gastric epithelial cells and gastric carcinoma cell lines,normal gastric tissue and gastric cancer tissues.Using si RNA transfection gastric cancer cells by q RT-PCR method to detect LINC00473 silence effects and changes of the Cell proliferation and migration.Using the method of Western blot to investigate the genes related to EMT after inhibiting LINC00473 expression(E-cadherin?N-cadherin?snail?vimentin and so on).The effects of LINC00473 on the proliferation and migration of gastric cancer cells were investigated by CCK-8 cell proliferation test,clone formation assay and Transwell migration test.Plasmids were used to construct the overexpression plasmid of LINC00473 and transfected into BGC-823 and MGC-803 gastric cancer cell lines.q RT-PCR was used to detect the efficiency of over-expression,to explore the changes in the expression of EMT related genes and to influence the cell proliferation and migration after over-expression.The cell apoptosis were analyzed by flow cytometry.Results: q RT-PCR showed that the expression of LINC00473 was higher in the gastric epithelial cell lines,and the expression was different in different gastric cancer cell lines.Further analysis showed that the m RNA level of LINC00473 was lower in BGC-823 and MGC-803 gastric cancer cell lines and relatively high in SGC-7901 and HGC-27 gastric cancer cell lines.After the overexpression of LINC00473 in gastric cancer cell line BGC-823 and MGC-803,the expression of EMT related protein expression was manifested as the increase of E-cadherin expression,the decrease of the expression of N-cadherin,snail,vimentin,and the inhibition of the transformation of epithelial mesenchymal transition(epithelial-mesenchymal transition,EMT);related functional tests also found that in BGC-823,after overexpression of LINC00473 in the MGC-803 cell line,the CCK-8 experiment found that the OD450 value was lower than the control group,and the cell clone formed was smaller and thiner than the control group.The Transwell experiment found that the number of migratory membranes decreased obviously,inhibited the proliferation and migration of gastric cancer cells,and RNAi interference inhibited LINC00473 in SGC-7901 and HGC-27 gastric cancer cells.After the expression,the expression of EMT related protein was changed,the specific expression changed to E-cadherin,N-cadherin,snail and vimentin,induced epithelial mesenchymal transition(epithelial-mesenchymal transition,EMT),the number of clones in the flat clones increased and the CCK-8 experiment found that OD450 value was higher than the control group,Tran.Swell experiments showed that the number of migratory membranes increased significantly,and the proliferation and migration ability of cells increased to a certain extent.The results of apoptosis after interference showed that LINC00473 might inhibit apoptosis.Conclusions: LINC00473 was low expressed in tumor tissues.LINC00473 can promote the apoptosis,inhibit the occurrence of EMT and inhibit the proliferation and migration of gastric cancer cells.Therefore,LINC00473 will be a promising marker for the diagnosis of clinical gastric cancer and provides a new way of thinking for the treatment of gastric cancer.
