Objective(1)Study on the identification of Mongolian medicine Roudoukou-8 San;(2)Study on the fingerprinting of Mongolian medicine Roudoukou-8 San;(3)Establish a multicomponent quantitative analysis method of Mongolian medicine Roudoukou-8 San.Methods(1)TLC is used for the identification of nutmeg,costustoot,agarwood,clove,inula japonica and choerospondiatis fructus;(2)Dehydrocostus lactone as reference peak,HPLC is used to study the fingerprints of 10 batches of Roudoukou-8 San;(3)HPLC is used to establish the relative correction factors of ellagic acid,costunolide,dehydrocostus lactone and dehydrodiiso-eugenol which using eugenol as internal reference,the contents of 5 components in Roudoukou-8 San are measured by QAMS and ESM,and compare the results of the two methods,investigate the effects of different instruments and different columns on the relative correction factors.Results(1)TLC method for nutmeg and costustoot is established;(2)A total of 10 common patterns of fingerprinting of Roudoukou-8 San are established,24 common peaks are identified,and 8 peaks are identified by comparison with the reference substance;The similarity of 10 batches of Roudoukou-8 San fingerprints is compared with 0.9,and the 10 batches are divided into 2 groups by cluster analysis.(3)Establish the QAMS for content the determination of ellagic acid,eugenol,costunolide,dehydroroma lactone and dehydrodiisobutanol in Roudoukou-8 San,the relative correction factors for each component relative to eugenol are 12.5289,0.8236,1.5248,4.8877;QAMS and external standard is evaluated by relative error,the results shows that the relative errors between the two methods are less than 2%,indicating that there is little difference between the two methods;RSD of the relative correction factors on different instruments and on different columns are less than0.4%,indicating that changes in instrumentation and columns have less effect on the relativecorrection factors.Conclusions In this paper,the TLC identification method established are used as reference medicine,and are highly specific and can be used for the qualitative identification of Roudoukou-8 San;The established QAMS and HPLC fingerprinting method are simple,reliable,accurate and stable,which can provide reference for the quality evaluation of Roudoukou-8 San.
