Objective Through the observation of Eerdun-wurile pre reperfusion injury treatment on myocardial ischemia(MIRI)in a rat model of inflammatory factor TNFalpha,IL-6 beta,IL-1,MMP2,MMP9 and mRNA expression levels,and the impact on the area of myocardial infarction,the pretreatment of the MIRI protection mechanism.Methods The 60 adult male SD rats were randomly divided into 6 groups: sham operation group,model group,Danshen Pill group,Eerdun-wurile low dose group,Eerdun-wurile middle dose group,high dose of Eerdun-wurile group.Each group according to the requirements of administration,dosage: Compound Danshen dripping pill group(0.5g/kg),Eerdun-wurile,low.The high dose group(0.3,0.6,1.2g/kg),sham operation group and model group were given normal saline,rats were administered volume 10ml/kg,2 times a day.The continuous administration of Ig for 2 weeks,12 hours after the last administration,the left coronary artery ligation in rats anterior descending(LAD)30 min,reperfusion 120 preparation of min of acute MIRI model,after modeling successfully killed.Observed the following indicators: 1 using Evans Blue-TTC double staining for each rat myocardial infarction area of.2.Elisa method was used to detect the concentration of inflammatory cytokines TNF-?,IL-6,IL-1? in serum of each group.3.Real-Time qPCR method was used to detect the expression of TNF-?,IL-6,IL-1?,MMP2,MMP9 in myocardial tissue of mRNA.Results 1.Evans Blue-TTC staining showed that compared with the model group,Eerdun-wurile group and Danshen Pill group could reduce myocardial infarct size in rats was significantly reduced(P< 0.05),Eerdun-wurile high dose group and Danshen group,no significant difference(P > 0.05),but the effect of high dose group is better;2.Elisa test results show that: the model group serum TNF-? ? IL-6 ? IL-1? was significantly higher than that of sham operation group;each group compared with themodel group,TNF-?? IL-6?IL-1? content were decreased(P < 0.01);compared with Danshen Pill group,high dose of Eerdun-wurile group of IL-6,has the advantage of reducing IL-1?(P< 0.01),while the TNF-? drop There was no statistical difference(P > 0.05);3.Real-Time qPCR analysis showed that Each treatment group compared with the model group,MMP2,MMP9,TNF-??IL-6?IL-1? and mRNA were reduced to varying degrees(P < 0.05),can inhibit the expression of inflammatory MIRI in a rat model of myocardial tissue reaction,the overall effect of high dose of Eerdun-wurile group than other groups,and Eerdun-wurile groups there is dose-response relationship.Conclusion 1 Eerdun-wurile can reduce myocardial infarction area and ultimately the role of protecting the MIRI.2.Eerdun-wurile by inhibiting to control the inflammation,protecting the MIRI.
