Objectives To investigate the expression and correlation of m TOR,HIF-1? and IL-17 in endometrial carcinoma,and to analyze its clinical significance.Methods 44 cases with endometrial carcinoma were selected who were treated by surgery in Tangshan maternal and child care service centre hospital during 1st April,2016 to end of Oct,2017 as the study team.At the same time(uterine myoma or uterine adenomyosis)line full hysterectomy or clinical shave,hysteroscopy biopsy of 33 patients with normal endometrium,endometrial hyperplasia patients 21 cases,as control group.In study group at the age of 37 ~ 65 years,including 24 cases without menopause and 20 cases of menopause.40 cases were in period I and 4 cases in III,no II and IV period cases,clinical tissue specimens for precious,did not get rid of a 4 cases III period its muscular infiltration were ?1/2 muscular wall,so included in the Ib period to statistical analysis,did not do the comparison of the pathological staging.No muscle infiltration or muscle layer infiltration <1/2 wall of 28 cases,muscular infiltration ?1/2 muscular wall 16 cases;High differentiation(G1)22 cases,middle and low differentiation(G2~3)22 cases.Sample endometrial adenocarcinoma of 31 cases of endometrial adenocarcinoma with squamous differentiation in 11 cases,2 cases of endometrial serous carcinoma.Control group age 38 to 63 years,including 33 cases without menopause and 21 cases of menopause;33 patients with normal endometrium and 21 cases of endometrial hyperplasia.Postoperation with half an hour,to collect the fresh endometrial tissue removed by surgery or operation,putting in frozen sealing tube into the-80? ultra-low temperature frozen storage to standby,the corresponding wax block was taken from the pathology department.First,to detect the expression of m TOR,HIF-1? and IL-17 in the endometrium of the study team and the control group by immunohistochemistry.Then,19 cases of tissue samples of m TOR,HIF-1? and IL-17 high expression in the immunohistochemistry were screened,and were divided into four groups according to the degree of infiltration of muscle layer and differentiation level,and the Western blot quantitative detection was conducted.Image J analysis software was used to determine the expression level of gray value determination protein.Last,statistical analysis was performed.Results 1 Control group of normal endometrium,endometrial hyperplasia in patients with age(52.7±4.2 years,54.5±5.1 years)and menopause rate(60.6%,61.9%)and the study team with age(55.4±6.1 years)and compared with postmenopausal rate(54.5%),there was not statistically significant difference(P>0.05).2 The high positive expression rate of m TOR,HIF-1? and IL-17 in normal endometrium,endometrial hyperplasia and endometrial cancer(30.3%,57.1%,63.6%;30.3%,52.4%,72.7%;18.2%,33.3%,52.3%),all showed an upward trend,and the difference was statistically significant(P<0.05).3 In endometrial carcinoma,the high positive expression rates(87.5%,93.8%,75.0%)of m TOR,HIF-1? and IL-17 in the muscular layer infiltration 1/2 or more of the wall were higher than that(50.0%,60.7%,39.3%)of no or muscular infiltration <1/2,the difference was statistically significant(P<0.05).The high positive expression rates(86.4%,86.4%)of m TOR and HIF-1? in middle and low differentiation were higher than that(40.9%,59.1%)of the high differentiation,the difference was statistically significant(P<0.05).The high positive expression rate(68.2%)of IL-17 was also higher than that of the highly differentiated(36.4%),but the difference was not statistically significant(P>0.05).There was not statistically significant difference between their expression and pathological types,and menopause or not too(P>0.05).4 Western blot detection of quantitativeanalysis,m TOR,HIF-1? and IL-17 in the muscle layer infiltration of 1/2,of grey value(1.214±0.127,1.343±0.178,0.997±0.034)was greater than no muscular infiltration or muscle layer infiltration <1/2 of grey value(1.008±0.072,0.918±0.167,0.754±0.054),the difference was statistically significant(P<0.05).The grayscale value of m TOR and HIF-1? in the middle~ low differentiation(1.145±0.139,1.194±0.241)was greater than that of the high-differentiator(0.997±0.105,0.911±0.277),and the difference was statistically significant(P<0.05).The grayscale value of IL-17(0.873±0.122)was greater than that of the highly differentiated(0.858±0.174),but the difference was not statistically significant(P>0.05).5 m TOR,HIF-1? and IL-17 expression in endometrial carcinoma tissues were positively correlated(r=0.386,0.507,0.437),and they were statistically significant(P=0.016,0.001,0.006,P<0.05).Conclusions 1 m TOR,HIF-1? and IL-17 were highly expressed in endometrial carcinoma,and all three were positively correlated in endometrial carcinoma.2 The expression level of m TOR and HIF-1? in endometrial carcinoma was increased with the increase of the infiltration of the muscle layer and the histological grade.The expression level of IL-17 was increased with the deepening of the infiltration of the muscular layer.
