Kelp is a phytoplankton of the brown algae family.The main bioactive components are polysaccharides and bioactive peptides.Alginic acid is the most abundant polysaccharide and is not easily absorbed by the body.It is often processed into alginate.According to reports in the literature,kelp has biological activities such as hypoglycemic,hypolipidemic,and antioxidation.The potassium alginate researched in this paper is divided into high-polymerization potassium alginate and low-polymerization alginate according to the degree of polymerization.Bioactive peptides are small-molecule peptides?hydrolysates?produced after enzymolysis of kelp proteins.Previous work has investigated the hypoglycemic effect of low-poly alginate potassium,low-poly alginate potassium:kelp hydrolysates?3:1?,high poly alginate:yam polysaccharides:polysaccharides?3:1:1?.Efficacy,but the ingestion of low-grade potassium alginate alone can cause excessive potassium intake in the body and cause side effects.Therefore,based on the results of the previous work,this article studied on hypoglycemic,hypolipidemic,and antioxidant effects of diabetes mellitus rats by using high-poly alginate potassium,kelp hydrolysates,and low-poly alginic potassium:yam polysaccharide:lycium barbarum polysaccharide?3:1:1??hereina fter referred to as formula?.First of all,the raw materials were tested:the content of polysaccharide in high-poly alginate potassium was 72.43%;the protein content in the hydrolyzate was 30.56%;the total polysaccharide content in the formula was 79.55%.Secondly,the inhibitory effects of?-glucosidase and?-amylase on intestinal digestive enzymes were examined.The results showed that the IC50 value of high-poly alginate potassium was 5.32 mg/mL and 9.84 mg/mL,compared with the positive control group?carbohydrate?,they increased by 29.88%and 40.65%,respectively;the IC50 values of hydrolysates from seaweed were 8.24 mg/mL and 16.76 mg/mL,which were 54.73%and65.15%higher than those of positive controls,respectively;It was 5.18 mg/mL and 10.91mg/mL,which was increased by 27.99%and 46.47%,respectively,compared with the positive control group.The inhibitory rate of the kelp hydrolyzates was positively correlated with the concentration of the formula.Finally,SD diabetic rats were selected to examine the auxiliary hypoglycemic function and divided into 7 groups:normal control group,toxicology group,model control group,positive control group?metformin??83.3 mg/kg·d?,high,medium,low In the dose group?1500,500,250 mg/kg·d?,after 4 weeks of gavage administration,the results showed that high-poly alginate potassium,kelp hydrolyzates,and formula can improve weight loss caused by diabetes and can be effectively controlled.Blood glucose levels;high-dose group,high-dose potassium polyoxalate group,and kelp hydrolyzates medium dose group can effectively improve glucose tolerance?OGTT?,increase serum insulin?INS?levels,and reduce glycated serum protein?GSP?content,which lowers blood glucose,increases superoxide dismutase?SOD?activity,decreases malondialdehyde?MDA?content,improves antioxidant capacity,and reduces serum total cholesterol?TC?and triglyceride?TG?levels,with auxiliary hypolipidemic effect.The prescription can effectively improve the number of pancreatic islets,repair cells,and reduce hyperlipidemia complications by reducing hepatocyte fat vacuoles,cell deformation,and edema.The toxicological experiments in rats of high poly alginate potassium,kelp hydrolysates,and formula showed that 30 times human dose did not show abnormalities in normal rats.In summary,this study shows that high-poly alginate potassium,kelp hydrolysates,and formula have the auxiliary hypoglycemic,antioxidant and hypolipidemic effect through improving OGTT,increasing serum INS,SOD levels and reducing GSP,SOD,TC,TG levels.
