CEA⁺ Cells Promote Metastasis Of CEA^-/lo Cells In Colorectal Cancer

Objectives:To investigate whether the CEA⁺cells can promote the metastasis of CEA^-/locell through collective dissemination in colorectal cancer Methods:HE staining were used to detect whether exist of“collective dissemination”metastasis model in colorectal cancer;Immunofluorescence staining was used to detect the expression of CEA in CTC clusters;Liver metastasis models were established by injecting three kinds of clusters into the spleens of NOD/SCID mice.According to the numbers of liver metastasis,we can compare the metastasis ability of three kinds of clusters;Compare the migration and invasion ability of CEA^-/lo/lo clusters and CEA⁺CEA^-/loclusters by transwell assays;Inducing anoikis of CEA^-/lo/lo clusters and CEA⁺CEA^-/loclusters in vitro,compare the apoptosis cells of them by flow cytometry;compare the ability of forming orgnoids of CEA^-/lo/lo clusters and CEA⁺CEA^-/loclusters.Results:Microemboli'metastasis model can be found in colorectal cancer;There are different cell subgroups in circulating tumor cell clusters?According to the expression of CEA,cell clusters can be divided into CEA⁺clusters?CEA^-/lo/lo clusters and CEA⁺CEA^-/loclusters;Compare with CEA^-/lo/lo clusters,CEA⁺CEA^-/loclusters have high metastasis potential;Compare with CEA^-/lo/lo clusters,CEA⁺CEA^-/loclusters have high ability of invasion and migration;high ability to resist apoptosis;high potential to form orgnoids?Conclusion:CEA⁺cells promote metastasis of CEA^-/locells through collective dissemination in colorectal cancer...
