Objective: To assess the therapeutic effects of dendritic cells andcytokine-induced killer cell (DC-CIK) immunotherapy with FOLFOXregimen on advanced colorectal cancer with diffusely hepatic metastasis.Methods: The peripheral venous blood about50ml of patients wascollected by machine, and the peripheral blood mononuclear cells (PBMCs)were separated from the blood by the lymphocyte separation medium. ThePBMCs were activated to obtain a large number of DC-CIK cells withINF-α、IL-1and GM-CSF. Totally18patients with advanced colorectalcancer with diffuse hepatic metastasis from January1,2008to December31,2010were divided into2groups: Folfox group (group F) andDC+CIK+Folfox group (group DCF),9patients in each group. Patients ingroup F were treated by FOLFOX regimen according to the NCCN guidewhile those in group DCF were treated by DC-CIK cells immunotherapywith FOLFOX regimen. To statistical analysis the objective response rate, clinical benefit rate, the changes of immune function and tumormarkers and the postoperative2years survival rate in the groups.Results: At15days the DC-CIK cells proliferation reached to peak invitro, it times to be applied in clinical. In group F,0patient attainedcomplete remission (CR),2patients attained partial remission (PR) witheffective treatment rate of28.6%. In Group DCF,2patients attained CR,5patients attained PR with effective treatment rate of87.5%, significantlyhigher than that of group F (P=0.03).There was no significant difference inclinical benefit rate (CBR) between two groups (P=0.32). In the detectionof immunological indexes, CD4+, CD8+and value of CD4+/CD8+weresignificantly raised after the treatment in group DCF, with statisticallydifferences(P=0.03), while no significant difference in group F wasobserved. In group F, markers of gastrointestinal tumors were notsignificantly changed during the treatment, but were increased significantlyafter the treatment. In group DCF, markers of gastrointestinal tumors weredecreased slowly during the treatment and were kept stable after thetreatment. The2-year survival rate was0%in group F and was33.3%ingroup DCF, with significantly statistical differences (P=0.00).Conclusion: Culture of mononuclear lymphocytes cells in vitrocan obtain a large number of DC-CIK cells in a short period of time. At thesame time, DC-CIK cell immunotherapy with FOLFOX regimen canprolong the survival time in the treatment of advanced colorectal cancer with diffusely hepatic metastasis and deserves clinical spread andapplication.
