Cervical cancer is one of the most common tumors in women.Although the diagnosis and treatment of cervical cancer have achieved remarkable results in recent years,the prognosis of cervical cancer is not very good.The 5-year survival rate of cervical cancer patients is not very high.The lack of understanding of the pathogenesis of cervical cancer at the molecular level is crucial.Mirna can affect cell proliferation and migration by regulating the expression of its specific target gene.Studies have shown that mature miRNA bind specifically to the 3'utr region of the target gene mRNA with incomplete or complete base complementary pairing.And then inhibit the translation of target gene mRNA or promote the degradation of mRNA.The occurrence of many kinds of cancer generally involves the abnormal expression of miRNA,so miRNA can be used as a marker for the diagnosis of cancer.And it is closely related to the proliferation and metastasis of tumor,but the role of miR-137-3p in the carcinogenesis of cervical cancer is not clear.In this paper,we determined that the expression of miR-127-3p in cervical cancer was significantly higher than that in normal cervix by real-time qpcr.In order to detect the effect of miR-127-3p on proliferation and migration of cervical cancer cells,We overexpressed mir-127-3p in HeLa cells and found that overexpression of miR-127-3p could significantly inhibit the proliferation and migration of HeLa cells by cell proliferation and migration assay.Subsequently,we found that kif3 b is a potential target gene of miR-127-3p through the target gene prediction website of miRNA.Double luciferase assay and Western blot analysis showed that miR-127-3p could bind to the 3'utr region of KIF3 B and inhibit the expression of KIF3 B protein.At the same time,we found that the expression of KIF3 B in cervical cancer was significantly higher than that in normal cervical tissue.Through the correlation analysis,we found that the expression of miR-127-3p and KIF3 B was negatively correlated in tissues,which may be an important molecular mechanism of miR-127-3p regulating the proliferation and migration of cervical cancer cells.Moreover,over expression of KIF3 B in cervical cancer cells can effectively suppress miR-127-3p in cervical cancer cells decreased cell proliferation and migration.These results demonstrate that miR-127-3p is a potential markers of the diagnosis and treatment of cervical cancer and a potential target.
