tumors in women,Cervical Cancer(CC)has high mortality.Its development is a continuous process of pathological changes,from Cervical squamous epithelial atypical hyperplasia to develop into invasive Cancer often requires several ten years,and therefore,exploring the molecular mechanism of Cervical Cancer occurrence and development,to find new biomarkers,early detection and early treatment is important for improving the prognosis and survival rate of patients with Cervical Cancer.The purpose of this study is to analyze the influence of lncRNA H1FXAS1 expression and miR-324-3p expression on the clinical manifestations of cervical cancer.Methods:In this study,the expression and prognosis of lncRNA H1FX-AS1 in cervical cancer samples were analyzed by TCGA database and GEPIA website.Then,clinical specimens from 50 cervical cancer patients were collected,and the expressions of lncRNA H1FX-AS1 and miR-324-3p in tumor tissues and cells were detected by real-time quantitative fluorescence PCR.The relationship between the expression of lncRNA H1FXAS1 and miR-324-3p and tumor size,clinical stage of TMN and lymph node metastasis of cervical cancer was analyzed by chi-square test.Results:LncRNA H1FX-AS1 gene was down-regulation in cervical cancer tissue,and cervical cancer patients with low expression of LncRNA H1FX-AS 1 gene had larger tumors,higher malignant degree,more lymph node metastasis,and lower survival rate.miR-324-3p gene was higher expressed in cervical cancer tissues and cell lines,and was negatively correlated with lncRNA H1FX-AS1.Conclusion:LncRNA H1FX-AS1 may act an inhibitor of the occurrence and development of cervical cancer,and its low expression can predict poor prognosis,and the molecular mechanism of its fuction may be through targeting miR-324-3p.Background and objective:In recent years,long non-coding RNAs(lncRNAs)have become a focus of anti-cancer research.Although they do not encode proteins,they are also involved in a variety of regulatory processes such as cell cycle,proliferation,differentiation and apoptosis.In this study,two cervical cancer cell lines(Siha and HeLa)were used as models to explore the effects of lncRNA H1FX-AS1 on the proliferation,invasion,migration and apoptosis of cervical cancer cells.Methods:In this experiment,the changes of biological function of cells were observed by gene overexpression technology.The proliferation ability of cells was confirmed by CCK8 assay and clone formation assay.In terms of cell migration and invasion,scratch detection and Transwell assay are mainly used.Cell apoptosis was detected by flow cytometry.The volume and size of tumors in mice were measured.Results:LncRNA H1FX-AS1 down-regulates the expression level of miR-324-3p mRNA in cancer tissues through targeted binding with miR-324-3p,overexpression of lncRNA H1FX-AS1 gene can inhibit cell proliferation,clone formation,invasion and migration,and promote apoptosis.Upregulation of lncRNA H1FX-AS1 gene significantly reduced the size,volume,and weight of tumor in mice.Conclusion:LncRNA H1FX-AS1 affects the occurrence and development of cervical cancer by inhibiting cell proliferation,migration,invasion and promoting apoptosisBackground and objective:The ceRNA hypothesis is that mRNA transcription or lncRNA pseudogenetic transcription can be used as a kind of ceRNA to competitively bind with miRNA with the same MRE through miRNA response element(MRE),and then these ceRNAs could regulate the expression level of downstream genes and affect the function of cells.Recent studies have proved that lncRNAs can interact with miRNA act as a kind of ceRNA,and compete with mRNA expressed by other genes to bind the same miRNA.Thus,lncRNAs play a role in information exchange and biological function regulation in cells.In this study,lncRNA H1FX-AS1 was selected as the research object,and the molecular mechanism of lncRNA H1FX-AS1 as a ceRNA regulating the biological function of cervical cancer cells was further explored.Methods:The binding sites and potential target genes of miR-324-3p were predicted by bioinformatics website.The direct interaction between lncRNA H1FX-AS1 and the miRNA and the interaction between miR-324-3p and DACT1 were confirmed by double luciferase reporting assay,RNA immunoprecipitation assay,gene overexpression or knockdown assay.Western blot assay was used to detect the protein expression.Rescue assays examinec changes in cell phenotypes when different genes were overexpressed or knocked down.Results:LncRNA H1FX-AS1 can directly bind to miR-324-3p and inhibit the mRNA level of miR-324-3p.miR-324-3p can directly bind to DACT1 and inhibit the expression of DACT1 at the transcriptional and translation levels.LncRNA H1FX-ASl inhibits the proliferation,migration and invasion of cervical cancer cells and promotes apoptosis by silencing miR-324-3p and then upregulating the expression of DACT1 in cervical cancer cells.Conclusions:LncRNA H1FX-AS1 gene can competitively bind miRNA-324-3p to induce the release and up-regulation of miRNA-324-3p target gene DACT1 in cervical cancer cells,play an
