?Background? Esophageal cancer is one of the most common malignant tumors in the digestive system.Endoscopic esophagus carotid stenting is the simplest and most effective palliative treatment for patients with dysphagia.Currently,the self-expandable stent progress into two kinds of coated stents: with/without coated,but they both rely on the effect of physical expansion,to treat the esophagus narrow ailment,but cannot keep the tumor process from development,and cannot solve the restenosis isuue during tumor regrowth.Therefore,the research area of esophageal lumen minimally invasive treatment brings the forefront of the tumor treatment as well as efficiently and effectively to relieve dysphagia.To reduce the repetition of treatment,increase the patient's alive quality as well as elongate his/her survivorship.Presently,the dual function completed by succeed in developing the gold nanoshell coated esophageal stent,to improve the simultaneously operations on physical support and the lumen malignant lesions treatment.The effect of the gold nanoshell coated esophageal stent is nanometer gold stuff with favourable efficiency of thermal through in situ reduction surface plating on the surface of metal stents,by reducing the properties of polymer with high polymer nano gold plated film technology.Therefore,increase the function of heating flowmeter based on the previous cure of traditional physical bracket support.The rationale of the light and heat therapy is to elevate the surface temperature of nano gold so that to kill and wound the surrounding tissue.?Aim? The major idea of the study is to open up a new research field of the gold nanoshell coated esophageal stent effects on esophageal cancer: the safety and infrared thermotherapy.To provide the consultation valuation for esophageal cancer clinical application.?Methods? 1?Through the effect of electron microscope gold nanoshell for cell morphology and structure,CCK 8 experimental detection cell vitality,flow cytometry instrument to detect cell apoptosis and toxicity experiment,in order to prove the biological safety of gold nanoshell coated esophageal stent.At the same time,to relieve dysphagia by the effect of the physical support of the gold nanoshell coated esophageal stent,which was verified by the esophageal stenosis model.2?To prove the gold nanoshell coated esophageal stent was treated by using NIR laser irradiation.We use the thermal conversion efficiency and stability of gold nanoshell coated esophageal stent,and we observed them by infrared thermal imager at different power of 808 nm near infrared laser.?Results? 1?The effect of gold nanoshell on cell morphology and structure,cell viability,cell apoptosis and toxicity in vivo proved that the gold nanoshell coated esophageal stent has well biocompatibility.It is indicated that the gold nanoshell coated esophageal stent is secure and non-toxic.At the same time,through internal experiments,it was verified that the gold nanoshell coated esophageal stent has the physical support effect.2?The nanoparticles can be converted to heat by a near-infrared laser at 808 nm,and photothermal transformation is proved to be stable.The excellent stability of the photothermal conversion of the gold nanoshell coated esophageal stent provides an opportunity for the clinical application of this kind of functional stent.By in vitro tumor-burdened bone cells,in vitro esophageal and nude mice model in body irradiation experiments,proved that gold nanoshell coated esophageal stent by using NIR laser thermal effect is produced to melt the surrounding tissue.These results further confir that the gold nanoshell coated esophageal stent can effectively treat cancer,which is helpful for the local accurate ablation of tumor in the future application.?Conclusions? 1?The gold nanoshell coated esophageal stent is safe and non-toxic.2?The gold nanoshell coated esophageal stent has the physical support function to relieve dysphagia.3?The gold nanoshell coated esophageal stent can produce photothermal conversion through 808 nm near-infrared laser irradiation,and the thermal transformation is stable,resulting in thermal energy that can destroy the surrounding tissues and play the role of infrared thermotherapy.
