With the dramatic increase in the quality of life in contemporary society,it is extremely common in middle-aged and older adults that unbalanced diet leads to diabetes,which can cause a series of complication.Western medicine has a rapid and obvious hypoglycemic effect,but hypoglycaemic drugs?metformin,etc.?are chemical drugs and have large toxic and side effects.Therefore,there is an urgent need for some organic substances with very small toxic and side effects to slowly replace western medicine.Therefore,the appropriate dose with low toxicity,strong antioxidant capacity and promoted growth were selected after conducting toxicological safety evaluations on several kinds of yeast selenium,which were used to study the thypoglycemic effect of hyperglycemic model mice.The following are the main research methods and results:1.Production of yeast selenium.After sterilizing the biological fermentor and potato dextrose agar?YPD?medium,the fermentation conditions were adjusted and controlled.The seed liquid was added to the fermenter by 5%of the total amount of the fermentation broth.The next step was that sodium selenite was added to the fermenter in a batchwise manner,and yeast selenium was produced by fermentation for a certain period of time.In this experiment,5 kinds of yeast selenium were prepared according to the selenium addition and the fermentation time.2.Acute Toxicity experiment.The five kinds of yeast selenium were disposable gavaged to normal Kunming mice according to the modified Karber's method,and calculating the half lethal dose.The results showed that one kind of the yeast selenium's LD500 was 33.33 mg/kg,which was highly toxic.Therefore,the other four kinds of yeast selenium were used for subsequent tests.3.Subchronic toxicity experiment.130 male Kunming mice were randomly divided into 13 groups:control group and 4 kinds of yeast selenium in high,medium and low dose groups,which were continuously gavaged for30 days.The sub-chronic toxicity evaluation was performed using body weight,food intake,organ coefficient,and in vivo antioxidant index.The medium dose group of No.4 yeast selenium and the low dose group of No.5yeast selenium had good growth performance,high food utilization rate and less oxidative damage in vivo.so the two kinds of yeast selenium would be evaluated for accumulation in the next step.4.Accumulating experiment.40 healthy male Kunming mice were randomly divided into 4 groups:control group,the parallel group,No.4 and No.5 yeast selenium feeding groups,which were gavaged by dose escalation method.Accumulation evaluation was based on the accumulation coefficient,body weight,organ coefficient and residual selenium in the body.The accumulation coefficient of No.4 yeast selenium was 4.76,which had mild accumulation.5.Hypoglycemic experiment.30 male healthy Kunming mice were randomly divided into 3 groups and used to the test of the effect of yeast selenium on normal mice.80 high-glycemic model mice were randomly divided into 8 groups and used to the test of the Hypoglycemic effect of yeast selenium on hyperglycemic mice.The results showed that yeast selenium needed much time to play the hypoglycemic effect than metformin,and the lower dose yeast selenium group had no significant hypoglycemic effect.The optimal intake dose of playing hypoglycemic effect remains to be further studied.
