Artemisia argyi is a dry leaf of mugwort,which is one of the common medicines in Chinese medicine.The Dragon Boat Festival has the customs of hanging mugwort,drinking wine and so on;In recent years,moxibustion has become more and more popular,and the volatile components and their corresponding physiological activities of Artemisia argyi are attracting more and more attention;Hyperuricemia,as a disease with high incidence,is also widely concerned,chemical drugs have a great effect on the human body,it is particularly important to develop natural products to treat hyperuricemia.So,It is of great significance for its application and new product development to study the chemical composition of Artemisia argyi and its effect on Hyperuricemia.Based on the Artemisia argyi from huozhou,Shanxi Province as the research object,studied its conditions of enrichment,separation and detection of volatile components systematically,established a new method with HS-SPME-GC-MS to analysis and identify its volatile before and after burning,at the same time the inhibition of Xanthine Oxidase,antioxidation on renal function and liver XOD activity in mice with hyperuricemia were studied.The main research contents and conclusions were as follows:(1)Established a method to analysis volatile components of Artemisia argyiThe influencing factors of the separation and enrichment of the volatile components from Artemisia argyi were investigated systematically,and the HS-SPME-GC-MS analysis method was established.The optimal extraction performance was achieved under the following conditions: 50/30 ?m poly two vinyl benzene/carbon molecular sieve/poly two methyl siloxane(DVB/CAR/PDMS)solid-phase micro extraction head,0.8 g of sample with extracting at 75 ? for 50 mins,30 min equilibrium time and 4 min desorption time.GC/MS conditions: HP-5MS column(30.0m×250?m×0.25?m),inlet temperature 250?,split injection,split ratio 5:1.(2)Analysis and identification of Artemisia argyi volatile components before and after burningMass spectrometry and retention index were used to identify the volatile components of Artemisia argyi before and after burning,using area normalization method to quantify the relative percentage of each component.A total of 196 compounds were isolated in Artemisia argyi before burning by mass spectrum analysis and retention index,and the structure of 132 in them was identified,accounting for 94.00% of the total volatile substances.The main volatile components are 3-aminopyrazole,eucalyptol,?-cadinene,a total of 143 compounds were isolated in Artemisia argyi after burning,and the structure of 104 in them was identified,accounting for 88.22% of the total volatile substances.The main volatile components are phenol,acetic acid,indole.(3)Study on the internal,external of inhibitory effect on hyperuricemia and anti-oxidative propertiesThe total flavonoids and total phenols,inhibitory effect on XOD,and antioxidant activity in the extracts of Artemisia argyi by distilled water,anhydrous ethanol with ultrasonic assisted and traditional decoction were measured.Oteracil potassium was selected as the inducer of hyperuricemia mice,and the serum uric acid(UA),creatinine(Scr),urea nitrogen level(BUN)and liver XOD activity were determined by the ELISA.The results showed that the total flavonoids and total phenolics of the alcohol extract are higher,and the effect of XOD and free radical scavenging was better than others,the levels of UA,Scr,and BUN in hyperuricemia mice have reduced to some extent,and they have inhibitory effects on liver XOD activity.
