Forsythia suspense,the air-dried fruit of Forsythia suspense?Thunb.?Vahl.,is a traditional Chinese medicine and used commonly,with heat clearing and detoxicating,detumescence,dispelling wind and heat effect,its fruit is used as medicine.Modern research shows that Forsythia suspense has antioxidative,bacteriostatic,anti-inflammatory and other pharmacological effects,and these pharmacological actions are closely related to chemical constituents of Forsythia suspense.In recent years,the increasing number of diseases is caused by environmental pollution,bad eating habits and other factors,due to increasing of the demand for traditional Chinese medicine.At the same time,there are some fake and shoddy drugs,which seriously threaten human health.With the development of social economy,the methods of quality standard and content determination of traditional Chinese medicine have gradually produced some limitations,and the method of biological activity determination has been put forward.Therefore,it is of great significance to explore the biological activity determination method of Forsythia suspense.Based on the pharmacological action of Green Fructus Forsythia in clearing heat and detoxification,the antioxidant,bacteriostasis and anti-inflammatory activity of this subject were studied.The samples of different Green Fructus Forsythia,different extracts of Green Fructus Forsythia and different plant organs were analyzed by HPLC,determined of total phenolic content and evaluated of antioxidant and antimicrobial and anti-inflammatory activity,and the correlation between total phenolics and antioxidant activity,total phenolic and bacteriostatic activity,antioxidant activity and bacteriostatic activity,composition and antioxidant activity,composition and bacteriostatic activity,antioxidant activity and anti-inflammatory activity,bacteriostatic activity and anti-inflammatory activity is analyzed with the bivariate correlation analysis,to laing the foundation for the study of biological activity for Green Fructus Forsythia.The specific contents are as follows:1.Taking antioxidant and bacteriostatic activity as screening indexes,extracts of Green Fructus Forsythia were selected from 0%,20%,40%,60%,80%and 95%ethanol,and the best extraction solvent was selected.At the same time,the fingerprint analysis was carried out.The experimental results showed that the content of the main components for 60%ethanol extract of Green Fructus Forsythia were the highest,and the antioxidant activity and bacteriostatic activity were the strongest,that is,60%ethanol extract of Green Fructus Forsythia had the best effect.2.30 batches of Green Fructus Forsythia were extracted by 60%ethanol,and were analyzed by HPLC,and determined of total phenolic content and antioxidant activity and bacteriostatic activity,and the correlation between total phenolics and antioxidant activity,total phenolic and bacteriostatic activity,antioxidant activity and bacteriostatic activity,composition and antioxidant activity,composition and bacteriostatic activity is analyzed with the bivariate correlation analysis.The results showed that the 30 batches of Green Fructus Forsythia all had antioxidant and bacteriostatic activities,while the total phenolic content had significant correlation with antioxidant activity and bacteriostatic activity.The correlation analysis showed that forsythoside I,forsythoside A,rutin,?+?-pinoresinol-?-D-glucoside,forsythin and so on were the antioxidative components,and the bacteriostatic activity constituents contained forsythoside I,forsythoside A and so on.3.The alcohol extract of Green Fructus Forsythia was preliminarily separated by middle low pressure preparative separation technology,and the antioxidation,bacteriostatic and anti-inflammatory activities of different elution sites?F1,F2,F3,F4,F5,F6?and crude extract?FF?were evaluated.The experimental results show that F2 component group with forsythiaside A and forsythiaside I were all the strongest for antioxidant,bacteriostatic and anti-inflammatory activity,that is the main active site of Green Fructus Forsythia.For antioxidant activity,phenylethanoid glycosides are the main material basis.For the bacteriostatic and anti-inflammatory activity,the activity of forsythoside A and forsythoside I were the strongest,so that forsythiaside I can be used as a quality control index of Forsythia suspense.In addition,the bivariate correlation analysis showed that the anti-inflammatory activity and antioxidant activity had a significant correlation.4.This chapter was to analyze the fingerprint spectrum,determine the contents of total phenol and total flavonoids,and evaluate antioxidant activity and bacteriostatic activity in Green Fructus Forsythia,Grown Fructus Forsythia,Forsythia suspensa root and Forsythia suspensa stem.Through comparison and analysis,it was found that all of them had inhibitory effect on Staphylococcus aureus and had good antioxidant capacity.Among them,Green Fructus Forsythia had the strongest antioxidant and bacteriostatic ability.In addition,Forsythia suspensa stem also showed stronger inhibitory ability to Staphylococcus aureus,and has potential utilization value.
