Objective:To investigate the expression of IncRNA-HEIH in coloretal cancer(CRC),and analysis the correlation of lncRNA-HEIH expression with the clinical pathological parameters of CRC patients.To investigate the impact of lncRNA-HEIH on cell proliferation,Apoptosis,cell cycle,migration and invasion ability of CRC cells.To reveal the function of lncRNA-HEIH in the occurrence and development of CRC and provide a new marker for diagnosis and treatment of CRC.Methods:84 CRC tissues and corresponding adjacent carcinoma tissues were selected and clinical pathological data were obtained.Real-time fluorescence quantitative polymerase chain reaction(RT-PCR)method was used to detected the expression of lncRNA-HEIH in CRC and corresponding adjacent carcinoma tissues.We analyze the relationship of the lncRNA-HEIH expression and the clinical pathological data of CRC patients.Further,we detect the influence of overall survival by lncRNA-HEIH using the Kaplan Meier method.RT-PCR method was used to detected the relative expression of lncRNA-HEIH in CRC cell lines(HT-29,SW480,HCT116 and LoVo)and an intestinal epithelial cell line(NCM460).CRC cells LoVo which expression of lncRNA-HEIH was the highest among all the CRC cells was interfered by lipsome in order to decrease the expression of lncRNA-HEIH,and we used RT-PCR method to detect the interference efficiency.After the expression of IncRNA-HEIH has been inhibited,the cell proliferation,apoptosis,migration and invasion ability of CRC cell LoVo were detected by CCK-8,flow cytometry,cell scratch test and Transwell methods.The expression of cell apoptosis related proteins including cleaved caspase3,cleaved caspase9 and bcl-2 were detected by western blot method after transfection.ResultsThe relative expression of lncRNA-HEIH in CRC tissues and the adjacent cancer tissues were 11.98±5.54 and 3.15 1.48.The relative expression of lncRNA-HEIH in CRC tissues was significantly higher than that it in the adjacent cancer tissues(P<0.05).The expression of lncRNA-HEIH in CRC tissue was significantly associated with the tumor size and TNM stages(P<0.05),and no significant associations were identified between lncRNA-HEIH expression and patient age,gender,tumor location,and tumor differentiation,and N classification(P>0.05).The Kaplan Meier curve showed the high IncRNA-HEIH expression group was obviously associated with shorter overall survival time(P<0.01).Multivariate analysis results showed high expression of lncRNA-HEIH was a significant independent prognostic factor for overall survival in CRC patients.The expression of lncRNA-HEIH in CRC cell lines HT-29,SW480,HCT116 and LoVo were 3.12±0.37,4.45±0.41,4.82±0.47 and 6.29±0.78,which were all significantly higher than that it in intestinal epithelial cell line NCM460(P<0.01).The LoVo cell exhibited the highest expression level among all the CRC cell lines.Compared to the control group,the cell proliferation,migration and invasion abilities of CRC cell were significantly decreased in lncRNA-HEIH inhibitor group(P<0.01).While the apoptosis rate in IncRNA-HEIH inhibitor groups were significantly higher than that in the control group(P<0.01).Cell cycle detection results showed CRC cell LoVo was blocked in G2/M period after the expression of lncRNA-HEIH inhibited.The expression of cell apoptosis related proteins cleaved caspase3 and cleaved caspase9 in LoVo were significantly increased after lncRNA-HEIH has been inhibited,while bcl-2 were significantly decreased(P<0.01).Conclusion:The expression of lncRNA-HEIH in CRC was significantly upgrading,and the high expression of lncRNA-HEIH was significantly associated with the tumor differentiation,TNM stages and the poor prognosis.Overexpression of lncRNA-HEIH promotes cell proliferation,migration and invasion abilities in CRC,and inhibit the cell apoptosis ability.
